外源基因在体内表达实验研究

摘要 目的:探讨Wistar大鼠舌肌是否具有摄取外源DNA并表达其携带基因的能力。方法:将质粒pSV40-β-gal扩增、纯化后溶于30% 蔗糖溶液中,取50 μg 质粒DNA 直接注射于Wistar大鼠舌肌中。1 周后用X-gal组织化学染色分析质粒DNA 在体内表达。结果:在Wistar大鼠舌肌中检查出报导基因产物β- 半乳糖苷酶的表达。结论:Wistar 大鼠舌肌具有摄取外源DNA并表达其携带的基因的能力,是一个理想的用来体内分析外源基因表达。 Objective:To study whether Wistar rat tongue has the ability to uptake foreign DNA and express its carrying reported gene.Methods:Plasmid pSV40 β gal was amplified,purified and dissolved in 30% sucrose.Fifty μg of the plasmid was injected into Wistar rat tongue.The reported gene E.coli LacZ product was detected using X gal staining after one week.Results:The reporter gene expression was foung in Wistar rat tongue muscle after plasmid DNA injection.Conclusion:Wistar rat tongue muscle could uptake foreign DNA and express its carrying reporter gene.Tongue is a good model to study the functions of certain gene regulatory elements in vivo.
