摘要
目的 :为探讨中药促神经生长的分子机理提供实验依据。方法 :采用 dd-PCR方法 ,从体外培养大鼠背根神经节细胞加中药组和不加中药组中获得两者的差异表达片段 ,并克隆测序分析。结果 :获得 7个差异条带 ,其中 2个条带的 c DNA序列分别与酪氨酸磷酸酶受体 ( PTPR) m RNA、促生长素抑制素受体 ( SSTR) m RNA的序列部分同源。结论 :在中药促神经生长过程中 。
Objective: To explore the molecular mechanism of the effect of Chinese herb upon nerve growth .Methods: The differential display of cDNA fragments had been obtained from two kinds of cultured cells from dorsal root ganglia - with Chinese herb and without Chinese herb by dd-PCR. Then, cDNA fragments were cloned and sequenced automatically. Results: Seven differential fragments were observed between the two kinds of cultured cells. Sequence of these fragments had been detected. Homology analysis with GenBank BLAST 2.0 showed that one shared some homology with PDPR, and the other had some homology with SSTR.Conclusion: Under the effect of Chinese herb on nerve growth, correlative gene displays differential expression.
