异种脱钙骨基质诱导异位骨髓形成的实验研究

摘要 目的 :开展诱导异位骨髓形成的研究 ,探讨骨髓形成及其调控机理。方法 :采用异种 (人 )脱钙骨基质粉 ,植入 Spraque-Dawley大鼠腹壁两侧皮下 ,建立异种脱钙骨基质粉诱导异位骨髓形成的动物实验模型。结果 :在诱导第 7天可见间充质细胞包绕并迁入骨基质性植入物的间隙 ,分化成原始软骨细胞、形成软骨组织。诱导第 13天 ,新生软骨组织进一步增加 ,成骨细胞、破骨细胞和血管位于其中 ,呈现软骨化骨现象 ,类骨样基质产生 ,成骨细胞下陷变为骨细胞。诱导第 17天 ,大多数软骨细胞集团内部腔隙融合并逐渐扩大形成初级骨髓组织。至 3 4天 ,原植入的脱钙骨基质大部分已被吸收并出现典型的骨髓组织 ,可见到大量髓细胞产生。在诱导的40、5 0、6 0天 ,诱导出的骨髓组织未见进一步增加 ,也未见明显萎缩。结论 :人、鼠骨诱导活性因子 ( BMP)具同源性 ,无种的特异性 ;诱导骨髓伴随软骨内骨化过程而产生 ,与骨组织的生成。 Objective:To develop the study of inducing ectopic bone marrow formation and to explore the mechanism of bone marrow formation and modulation.Methords:To establish an animal experimental model of ectopic bone marrow formation induced by heterogeneous demineralized bone matrix with transplanting the heterogeneous(human) demineralized bone matrix in Spraque-Dawley rat's bilateral abdominal subcutaneous tissue.Results:On the 7th day,mesenchymal cells enclosed the engrafted bone matrical transplant,migrated into the gap,differentiated to immature cartilage cell and formed cartilage tissue.On the 13th day,newborn cartilage tissue further increased,osteoblast,osteoclast and blood vessel located in the cavity,cartilagonous ossification occurred,formed analogous bony matrix,osteoblast differentiated to bone cell.On the 17th day,most of the innteral cavity of cartilage cells mass gradually merged,expanded and formed primary bone marrow.On the 34th day,most of the engrafted demineralized bone matrix were absorbed,typical bone marrow tissue appeared and a great quantity of myloid cells were seen.Neither further increase nor obvious atrophy of the induced bone marrow tissue was seen on the 40th,50th,60th day.Conclusion:Bone inducing active factor(BMP) of human and rat is homology.There is no heterogeneous.Induced bone marrow appeared with the procedure of cartilafonous ossification.It consists with the formation and development of bone tissue.
