培养的周围神经组织特殊三色染色法

THE SPECIAL METHOD TO CULTURE PERIPHERAL NERVE TISSUE BY TRICHROME STAINING

目的 :以特殊三角染色法显示培养的周围神经组织中轴索及雪旺氏细胞等结构。方法 :大鼠背根神经节体外培养后 ,用苏木素精、固绿 FCF、变色素 2 R及磷钨酸等配制三色染色液染色。结果 :神经轴索呈蓝绿色 ,雪旺氏细胞核为红色或紫红色 ,胞质为浅灰色。结论

Objective:To show neurites,Schwann's cell in culture of peripheral nerve tissues by special trichrome stain.Methods:The dorsal root ganglia of SD rat were cultured in vitro.Then,the cultured specimens were stained by special stain,which was composited of hematoxylin fast green FCF、chromotrope 2R and phosphotungstic acid.Results:The neurites were stained aquamarine blue.The nuclei of Schwann's cell were stained red of purplish red;meanwhile,the cytoplasm of the Schwann's cell was stained ashen.Conclusion:The special stain could offer differentiated colors between neurites and Schwann's cell in culture peripheral nerve tissue.
