摘要
目的:观察四逆汤及配伍药提取物对内皮细胞释放PGI2的影响:。方法采用家兔血管内皮细胞培养1,25I标记6-酮-PGF1α浓度的放射免疫方法,在低氧条件下,测定内皮细胞释放的前列环素代谢产物6-酮-PGF1α(6-keto-PGF1α)的含量。结果:与对照组比较,四逆汤、附子、甘草、干姜、附子配甘草、附子配干姜提取物12.5μg/ml和25μg/ml均能显著提高6-keto-PGF1α的含量,除甘草25μg/ml剂量组和附子配干姜组外,其它组6-keto-PGF1α的含量显著低于四逆汤组。单体提取物在10μg/ml剂量,与附子配伍的干姜单体中附子+5-羟基-1-(3-甲氧基4-羟基苯基)癸酮-3、附子+5-羟基-1-(3-甲氧基苯基)癸酮-3组作用较强。与干姜配伍的附子单体中干姜+苯甲酰中乌头碱、干姜+总脂肪酸酯生物碱组作用显著。结论:在病理状态下,四逆汤及配伍药提取物对内皮细胞释放PGI2表现出良好的配伍增效关系,与干姜有协同增效作用成分是从附子中提取的苯甲酰中乌头碱和总脂肪酸酯生物碱,与附子有协同作用的成分是从干姜中提取癸烷和癸酮。揭示了四逆汤配伍规律的药效作用物质基础。
Objective:To compare the effects of the extracts from Sini Decoction(四逆汤) and its compatibility herbs on prostacyclin(PGI2) release from rabbit vascular endothelial cells in vitro.Methods:the content of 6-keto-prostaglandin F1α(6-keto-PGF1α)in culture medium of rabbit vascular endothelial cells under hypoxia were measured with radioimmunoassay.Results:Compared with the control group,all the extracts of Sini Decoction(DRE),aconite(AE),licorice(LE),dried ginger(GE),aconite and licorice(ALE),aconite and dried ginger(AGE) at the dosages of 12.5 and 25μg/ml can increase significantly the content of 6-keto-PGF1αin culture medium of rabbit vascular endothelial cells.Except for LE group at the dosage of 25μg/ml and AGE group,the content of 6-keto-PGF1α of other groups were all decreased significantly compared with that of DRE corresponding dosage group.Among the compatibility of AE and GE monomers,AE+HMHD-3 and AE+HMD-3 at the dosage of 10μg/ml had the strongest effect of increasing PGI2 release from vascular endothelial cells.Among the compatibility of GE and AE monomers,GE+BM and GE+AFAE had the strongest effect of increasing PGI2 release from vascular endothelial cells.Conclusion:DRE and its compatibility herbs have cooperative synergism effect on rabbit vascular endothelial cell releasing 6-keto-PGF1α under pathologic condition.The components having synergetic effect with GE are benzoyl mesaconitine and total fatty acid ester alkaloids extracted from AE.The components having synergetic effect with AE are decane and decanone extracted from GE.This research reveals material basis of the DRE pharmacodynamics.
