摘要
目的:探讨含人工麝香血清对心肌细胞的保护作用。方法:制备含人工麝香血清,建立体外培养心肌细胞模型,应用血清剥夺法造成心肌细胞缺血后,给予不同剂量含药血清进行处理,观察心肌细胞形态、自发性搏动频率及培养液中MDA、LDH、NO含量的变化。结果:人工麝香1:2稀释组增加NO释放(P<0.05);人工麝香血清:空白血清(1:1)组减缓缺血损伤造成的心肌细胞自发性搏动频率下降程度(P<0.05),降低缺血所致的LDH活力的升高(P<0.05),增加NO的释放(P<0.01);未稀释的含人工麝香血清组降低缺血所致的LDH活力的升高(P<0.01)、MDA含量的增加程度(P<0.05),增加NO的释放(P<0.01)。结论:含人工麝香血清对体外培养的心肌细胞具有保护作用,可能通过其促进NO生成,减少LDH,MDA释放有关。
Objective:Explore contain artificial musk serum of myocardial cell protection.Method :Preparation contain artificial musk serum myocardial cells,establish the in vitro model was applied to cause myocardial cell serum deprive method after ischemia,Give different doses of medicated serum processing,the myocardial cells form,spontaneity pulsations frequency and medium MDA and LDH,NO content change.Result :Contain artificial musk chroma in serum group ischemia injury caused by slowing myocardial cells spontaneous pulsations frequency decrease degree(P 〈 0.05),reduce ischemia due to dynamic LDH increased(P 〈 0.05),increase the release of nitric oxide(NO)(P 〈 0.01),High concentrations of group of LDH caused by reducing ischemia dynamic increased(P 〈 0.01),the increase of the content of MDA degree(P 〈 0.05),increase the release of nitric oxide(NO)(P 〈 0.01),Low concentration increases the release of nitric oxide(NO) group(P 〈 0.05).Conclusion :Contain artificial musk serum myocardial cells in vitro cultured protectie effect,possibly through its promote nitric oxide formation,Reduce LDH,MDA release relevant.
