藕节炭活性部位止血机制的研究 被引量:16

Research on hemostatic mechanism of charred Nodus Nelumbinis Rhizomatis

摘要 目的:确定藕节炭止血作用的有效部位和成分的基础上,探讨有效部位及其成分的止血作用机制。方法:大鼠灌胃给药(10.8g/kg),连续给药1周,末次用药后1 h0,.3%戊巴比妥腹腔注射麻醉,腹主动脉采血,0.109 mol/L枸橼酸钠抗凝(1:9体积)3,000 rpm离心10 min,分离血浆。测定血浆凝血酶原时间(PT),活化部分凝血活酶时间(APTT),纤维蛋白原(FIB)及D-二聚体含量,血小板聚集功能及血液流变学指标。EDTA-Na2抗凝,3000rpm离心10 min,分离血浆,放免法测定血浆TXB2和6-Keto-PGF1a。结果:三个受试药物不同程度缩短凝血酶原时间和部分活化凝血酶原时间,增加血浆纤维蛋白原的含量,降低血浆中纤维蛋白降解产物的浓度;提高血浆血栓烷B2含量,降低6-酮-前列腺素F1a含量。血小板聚集功能试验可见三个给药组不同程度提高血小板聚集率,对各受试药物进行血液流变学主要指标观察,可见血液粘度不同程度增加,红细胞电泳时间不同程度延长。结论:藕节凝血作用靶点涉及到凝血、抗凝及其血栓形成的整个过程,并由此导致血流动力学的改变,起到凝血的功效。但其有效成分表白桦脂酸的凝血作用靶点少,可能只对凝血、抗凝和血栓形成过程中某些部位起作用。 Objective:To determine the role of an effective hemostatic Oujie(藕节) carbon parts and components,based on the effective parts and components of the hemostatic mechanism of action.Methods: Wistar rats,male and female in half,according to weight,gender balance randomly grouped continuous administration for 1 week after treatment last time 1 h,0.3% intraperitoneal injection of pentobarbital anesthesia,abdominal aortic blood,0.109 mol / L sodium citrate(1:9 volume),3000 rpm centrifugation 10 min,separated plasma.Determination of plasma prothrombin time(PT),activated partial thromboplastin time(APTT),fibrinogen(FIB) and D-dimer levels,platelet aggregation and blood rheology.EDTA-Na2 anticoagulant,3000rpm centrifuge 10 min,separated plasma was measured by radioimmunoassay in plasmaTXB2 and 6-Keto-PGF1a.Results: The three subjects in varying degrees of drug shortened prothrombin time and partial activation of prothrombin time and increased plasma fibrinogen levels,lower plasma concentrations of fibrin degradation products.Increase in plasma thromboxane B2 levels and lower 6-keto-prostaglandin F1a levels.Platelet aggregation function test can be seen to varying degrees in three treatment group increased platelet aggregation rate,drugs on various subjects Hemorheological observation,showing varying degrees of increased blood viscosity,erythrocyte electrophoresis time of the extension to varying degrees.Conclusion: The role of targets related to Oujie coagulation coagulation,anticoagulation and thrombosis of the whole process and the resultant hemodynamic changes play a clotting effect.But its active ingredient the role of coagulation in Table betulinic acid target small,probably only on blood coagulation,anticoagulation and thrombus formation play a role in some parts.
