仙茅醇提取物大鼠长期毒性试验研究

Research on long term toxicity of ethanol extracts of Curculigo orchioides Gaertn

目的:研究仙茅乙醇提取物重复给药对大鼠所产生的毒性作用和靶器官,毒性损害的可逆性,剂量-反应(效应)关系,并探讨可能的毒性作用机制。方法:设仙茅醇提物120g生药/kg、60g生药/kg3、0g生药/kg和空白对照组(等容量蒸馏水),每日灌胃给药1次,连续给药30天、90天以及停药恢复性观察15天。观察一般症状和体征、血液细胞学、血液生化学及肉眼、光镜和电镜病理学观察。结果:仙茅乙醇提取物灌胃给药30天后,未见明显异常;90天后1,20g/kg组血清BUN高于对照组(p<0.05)1,20、60、30g/kg组CREA高于对照组(p<0.01),1206、0g/kg组ALT高于对照组(p<0.05～0.01)。高、中、低剂量组雌、雄大鼠肝脏和肾脏器系数高于对照组(p<0.01),高、中剂量组雄性大鼠睾丸脏器指数高于对照组(p<0.05),中剂量组子宫脏器指数低于对照组(p<0.05)。光镜下仙茅醇提物大剂量组大鼠的主要脏器无明显改变,透射电镜下观察发现给药高剂量组大鼠肝、肾无明显改变,而雄性大鼠的睾丸及雌性大鼠的卵巢均呈现线粒体肿胀,空泡等病理改变。停药15天后观察,上述各项指标与对照组比较无显著差异。结论:仙茅乙醇提取物120g/kg长期反复给药,可能引起血清BUN、CREA及ALT升高,肝脏、肾脏、睾丸和卵巢系数增加,透射电镜可见睾丸和卵巢主要表现为线粒体肿胀,空泡变性等超微结构病理学改变。

Objective：Research the target organs and mechanism of action of ethanol extracts of Curculigo orchioides Gaertn.through the long term toxicity.Methods：Set the high dose（120g/kg）、medial dose（60g/kg）、low dose（30g/kg）and the control group,intragastric administration daily dosing 1 times,continuous dosing 30 days、90 days and withdrawal restorative observation 15 days.Observe the general symptoms,blood cytology and blood biochemistry,naked eye,light and electron microscopic observation pathology.Results：intragastric administration dosing after 30 days,there is no abnormal;90 days,the liver and kidney coefficients of high,medial and low dose are higher than the control group（P〈0.01）,the testis coefficient of high,medial dose are higher than the control group（P〈0.05）,the uterus coefficient of medial dose is lower than the control group（P〈0.05）;the BUN in blood serum of of high dose is higher than the control group（P〈0.05）,the CREA in blood serum of high,medial and low dose are higher than the control group（P〈0.01）,the ALT in blood serum of high,medial dose are higher than the control group（P〈0.050.01）.Under the light microscope,major organs have no changes;Under the electron microscope,the livers and kidneys of high dose have no changes,but the testis and the ovarian present the pathological changes as mitochondria swelling.drug withdrawal 15 days after,each index has no notable difference.Conclusions：Intragastric administration witrh Ethanol extracts of Curculigo orchioides Gaertn.with high dose and long term,may cause the pathological changes of testis of male rats and the ovarian of female rats as mitochondria swelling,and it can also change the liver and kidney functions.
