摘要
目的:研究槲芪散对肝癌前病变大鼠肝脏血流的影响,探讨槲芪散干预肝癌前病变的机制。方法:依据Solt-Farber程序建立肝癌前病变动物模型,应用高分辨率超声影像系统检测大鼠肝脏门静脉、肝静脉内径及血流速度;利用酶组织化学染色的方法检测大鼠肝脏组织能量代谢相关酶G-6-Pase葡萄糖-6-磷酸酶、SDH琥珀酸脱氢酶、ATPase三磷酸腺苷酶的活性。结果:肝癌前病变大鼠的门静脉内径及门静脉血流速度增加、肝静脉内径及肝静脉血流速度减少。经槲芪散干预的肝癌前病变大鼠的门静脉内径及门静脉血流速度减少、肝静脉内径及肝静脉血流增加。肝癌前病变大鼠能量代谢相关酶G-6-Pase葡萄糖-6-磷酸酶、SDH琥珀酸脱氢酶、ATPase三磷酸腺苷酶的活性发生变化,槲芪散对此变化有干预作用。结论:槲芪散可以减轻肝癌前病变大鼠肝脏的瘀血状态,促进肝脏糖异生及有氧氧化,改善肝脏能量代谢。
Objective:To investigate the effect of Huqisan on liver blood flow of the DEN-induced hepatic precancerosis rats and the mechanism of the effects of Huqisan on hepatic precancerosis.Methods :The hepatic precancerosis animal models were established according to the Solt-Farber procedure.The hepatic portal diameter and hepatic vein diameter and blood velocity were detected by the High Resolution Imaging Sys-tem;The activity of the energetic metabolism related enzyme such as G-6-Pase glucose-6-phosphatase and SDH succinatedehydrogenase and ATPase adenosine triphosphatase were detected by histochemical staining method.Results: The hepatic portal diameter and blood velocity in-creaced in the model group and decreaced in the treatment group;The hepatic vein diameter and blood velocity decreaced in the model group and increaced in the treatment group.The activity of G-6-Pase glucose-6-phosphatase and SDH succinatedehydrogenase and ATPase adeno-sine triphosphatase have changed both in the model and treatment groups.Conclusions: Huqisan can alleviate the state of liver congestion of the precancerosis rats and promote the liver glyconeogenesis and aerobic oxidation and improve the liver energy metabolism.
