抗纤复方药物血清对肝纤维化大鼠肝细胞生成胶原的影响

Effects of compound anti fibrosis prescription on collagen synthesis in hepatocytes of rat fibrotic liver

目的 为研究抗纤复方药物的抗肝纤维化的细胞机制。方法 免疫细胞化学染色法比较观察了原代培养正常和纤维肝肝细胞Ⅰ、Ⅲ、Ⅳ型胶原表达，并以血清药理学方法研究抗纤复方药物对纤维肝肝细胞生成胶原的影响。结果 正常肝细胞极少表达胶原，纤维肝肝细胞胶原表达增加，抗纤复方药物能有效抑制纤维肝的肝细胞生成胶原，其作用优于秋水仙碱。结论 抗纤复方药物能抑制纤维肝肝细胞合成胶原，而减少肝脏细胞外基质的沉积。

Objective To study the effect of compound anti fibrosis prescription (ACP) on collagen synthesis in hepatocytes of rats' fibrotic liver. Methods Expressions of Type Ⅰ, Ⅲ and Ⅳ collagen in primary cultured hepatocytes were detected by immunocytochemistry, and the effects of compound anti fibrosis prescription on collagen synthesis were studied with seropharmacological method. Results The results showed there was little collagen expression in hepatoeytes of normal rats, but the expression was increased in rats' fibrotic liver, ACP could effectively inhibit collagen synthesis in hepatocytes of the fibrotic liver and was superior to Colchicine. Conclusions The ACP can inhibit collagen synthesis in hepatocytes of rats' fibrotic liver and reduce the deposition of extracellular matrix in the liver.
