芎嗪对实验性糖尿病大鼠肾脏的保护作用

Renal protective effect of tetramethylpyrazine in diabetic rats

目的 探讨川芎嗪对实验大鼠糖尿病肾病的预防作用。方法 ＳＤ 大鼠３０ 只，分为正常对照组（ＮＣ）、糖尿病对照组（ＤＣ）和糖尿病治疗组（ＤＴ） 。各糖尿病组以四氧嘧啶腹腔内注射制成糖尿病模型。ＤＴ 组采用川芎嗪３０ｍｇ·ｋｇ－ １ 腹腔内注射，每周５ 次，共１２ｗｋ 。取肾脏行光镜和电镜检查，并行蛋白糖基化产物测定。统计学处理采用方差分析。结果 川芎嗪治疗可明显降低糖尿病大鼠肾组织中蛋白糖基化产物水平（ＤＴ与ＤＣ相比，Ｐ＜０ ．０１）；电镜则显示，ＤＣ肾小球基底膜明显不均匀增厚，而ＤＴ 大鼠病变较轻。结论 川芎嗪治疗可减慢实验大鼠ＤＮ 的进展。

Objective To search the prophylactic effect of tetramethylpyrazine (TMPZ) for diabetic nephropathy(DN).Methods Thirty SD rats were divided randomly into three groups:normal control group(NC),diabetes control group(DC),and diabetes treated group(DT).The diabetes model was established by intraperitoneal injection of alloxan.TMPZ (30mg·kg -1 ) was given to DT rats by intraperitoneal injection,5 times a week and for 12 weeks.The kidneys were processed for morphological examination with light and electron microscopes,and the glucosylation of protein(GP) in renal tissue was assayed.Results Compared with DC,TMPZ treatment could decrease significantly the level of GP in renal tissue in diabetic rats(P<0.01);the electron microscopic findings indicated that the glomerular basement membrane in DC rats was uneven thick significantly,and this abnormal changes were alleviated significantly in DT rats.Conclusion TMPZ could slow the progress of the DN in experimental diabetes rats.
