摘要
目的观察矮壮素(chlormequat chloride,CQ)对小鼠生殖功能的毒性作用。方法将144只成年无生育史清洁级ICR小鼠按体重随机分为4组,分别为对照(蒸馏水)组和低(20 mg/kg)、中(60 mg/kg)、高(180 mg/kg)剂量矮壮素染毒组,每组36只,雌∶雄为2∶1。采用灌胃方式进行染毒,染毒容量为0.02 ml/g,每天1次;雄鼠从交配前28 d连续染毒至交配结束,雌鼠从交配前14 d连续染毒至仔鼠断乳,仔鼠观察至断乳。测定亲代小鼠体重、相关脏器重量及其脏器系数和精子运动参数,测定断乳后子代小鼠体重、相关脏器重量及其脏器系数和出生数。结果在亲代小鼠中,与对照组比较,各剂量矮壮素染毒组雄性小鼠的体重下降(P<0.05),而雌性小鼠的体重无变化(P>0.05);高剂量矮壮素染毒组雄性小鼠的睾丸系数较高(P<0.05),各剂量矮壮素染毒组雄性小鼠的心脏、肝脏、脾脏、肺脏、胃、肾脏、附睾的脏器系数无变化(P>0.05),各剂量矮壮素染毒组雌性小鼠的心脏、肝脏、脾脏、肺脏、胃、肾脏、子宫、卵巢的脏器系数也无变化(P>0.05);中、高剂量矮壮素染毒组精子计数和高剂量矮壮素染毒组精子活动率较低(P<0.05)。在子代小鼠中,与对照组比较,高剂量矮壮素染毒组小鼠的出生数较低(P<0.05),中、高剂量矮壮素染毒组断乳后小鼠体重下降(P<0.05);各矮壮素染毒组小鼠心脏、肝脏、脾脏、肺脏、肾脏、胃的脏器系数无变化(P>0.05)。结论矮壮素可能具有生殖毒性和胚胎毒性。
Objective To observe the toxic effects of chlormequat chloride on the reproductive function of mice. Methods One hundred and forty-four ICR mice were randomly divided into 4 groups,36 in each group, female and male ratio was 2:1, including a control group (distilled water) and three experimental groups, 20, 60 and 180 mg/kg, 0.02 ml/g through gavage, once a day. For male mice, chlormequat chloride was administered for 28 days before mating,and for 14 days after mating; For female mice, the agent was administrated for 14 days before mating and continuing to the offsprings weaning. The offspring were observed until weaning. The mice and offspring were weighted, the organs coefficients and sperm kinematic parameters were determined, and the litter size was counted. Results Compared with the control group, the male mice body weight in experimental groups were lower (P〈0.05), the testis coefficient of high experimental group was higher (P〈0.05), the sperm counts in moderate and high dose groups were lower (P〈0.05), and the sperm motility rate in high dose group was lower (P〈0.05). The litter size in high dose group was significantly lower than that in control (P〈0.05). The weight of weaning offspring in moderate and high dose groups were significantly lower than that in control (P〈0.05). Conclusion Chlormequat chloride may have reproductive and embryonic toxicity to mice.
