摘要
目的研究香烟对肺部炎性介质IL-1、IL-6、IL-8分泌的影响。方法制备香烟烟雾提取物(CSE),用不同浓度的CSE(10%~100%)刺激肺泡Ⅱ型上皮细胞A549,以放射免疫法检测细胞培养上清液中IL-1、IL-6、11—8的水平。用不同吸烟剂量(低、中、高剂量组分别每天吸烟4、8、16支)制备动物吸烟模型,采用自主开发的吸烟机给烟12周,以放免法检测肺灌洗液(BALF)中IL-1、IL-6、IL-8的水平。结果各浓度CSE组IL-1水平与对照组相比,差异无统计学意义(P〉0.05),20%、50%、100%CSE组IL-6的水平明显升高,IL-8的水平下降,差异有统计学意义(P〈0.05);各吸烟剂量组IL-1的水平与对照组相比,差异无统计学意义(Jp〉O.05),低、中、高剂量组IL-6、IL-8的水平显著升高,差异有统计学意义(P〈0.05)。结论香烟会影响肺部炎性介质IL-6、IL-8的分泌。
Objective To investigate the effect of cigarette smoke on the expression of inflammatory mediators in lungs. Methods A549 cells were exposed to different concentrations of cigarette smoke extract (CSE), which was collected by using dimerhyl sulfoxide (DMSO) as absorbent. The level of IL-1,IL-6 and IL-8 was measured with radioimmunoassay. The animal models of smoking were established though different doses smoking exposure for 12 weeks. Inflammatory mediators in bronchoalveolar lavage fluid (BALF) were detected by radiation immune method. Results In cell experiment, no significant difference was seen in the expression of IL-1 among groups (P〉0.05). Compared with the control group, the IL-6 expressions in 20%, 50%,100% concentrations groups significantly increased (P〈0.05), the expression of IL-8 decreased (P〈0.05). In animal experiment, also, no significant difference was seen in the expression of IL-1 among groups (P〉0.05). The levels of IL-6 and IL-8 in BALF increased significantly in high-close group, moderate-dose group and low-dose group respectively compared with the control group (P〈0.05). Conclusion Cigarette smoke exposure can influence the expression of IL-6 and IL-8 in lungs.
