邻苯二甲酸二(2-乙基己基)酯对雌性小鼠子宫内膜雌激素受体α表达影响的研究 被引量:6

Effect of Di-(2-ethylhcxyl)phthalate on Expression of ERα in Endometrium Tissue of Female Mice

摘要 目的研究邻苯二甲酸二(2-乙基己基)酯(DEHP)对雌性小鼠子宫内膜雌激素受体(ER)α表达的影响。方法将健康成年清洁级雌性ICR小鼠60只随机分为4组,即对照(大豆油)组和低(125 mg/kg)、中(500 mg/kg)、高剂量(2 000 mg/kg)DEHP染毒组,每组15只。于早晨采用灌胃方式进行染毒,染毒容量为5 ml/kg,每天1次,每周6 d,连续染毒16周。染毒结束后,于动情间期处死小鼠,采用HE染色法观察子宫组织形态学变化,采用免疫组化染色法检测子宫内膜ERα的表达情况。结果各染毒组小鼠子宫组织固有层腺体数目减少,子宫内膜出现胞核假复层现象。与对照组比较,中、高剂量组子宫内膜免疫组化染色程度较强,范围较大。与高剂量DEHP染毒组比较,对照组和低、中剂量DEHP染毒组小鼠子宫内膜中平均光密度(mean density,MOD)值较高,差异均有统计学意义(P<0.05)。与对照组和低剂量DEHP染毒组比较,中、高剂量DEHP染毒组小鼠子宫内膜中积分光密度(integral optical densitiesI,OD)值较高,差异均有统计学意义(P<0.05)。且随着DEHP染毒剂量的升高,小鼠子宫内膜中MOD值和IOD值均呈上升趋势。结论 DEHP可促进小鼠子宫内膜ERα的表达。 Objective To determine the effects of di-(2-ethylhcxyl)phthalate (DEHP) on uterine morphology and the expression of ERα in the endometrium tissue of female mice. Methods Sixty ICR female mice were randomly divided into 4 groups, a negative control, three experimental groups, 15 in each,treated with DEHP at the doses of 125,500 and 2 000 mg/kg, through gavage,once a day,6 days a week,for 16 consecutive weeks. The mice were sacrificed during anoestrum to observe the change of uterine tissue morphology by HE staining,detect the expression of ERα in endometrium by immunohistochemistry. Results The nuclei of endometrium appeared pseudostratified phenomenon,and the number of glands in the lamina propria reduced in the experimental group. Degree and range of immunohistochemical staining in the moderate and high dose groups were stronger and larger compared with the control group in endometrium tissue. The mean density (MOD) of endometrial ERα in the high dose group was higher than that of the other groups; The integral optical densities (IOD) of endometrial ERα in the moderate and high dose groups were significantly higher than that of the control and low dose groups (P〈0.05). The MOD and IOD in the endometrium tissue of female mice gradually increased as the dose increased. Conclusion DEHP can increase ERα expression in the endometrium tissue of female mice.
