摘要 目的探讨Quil A对口蹄疫(Foot-and-mouth disease,FMD)疫苗的免疫增强作用。方法将O型FMDV抗体阴性的仔猪随机分为4组:1组首次免疫O型口蹄疫核酸苗pVIRIL18P1,第2次免疫重组鸡痘苗vUTAL3CP1;2组首次免疫Quil A+O型口蹄疫核酸苗pVIRIL18P1,第2次免疫重组鸡痘苗vUTAL3CP1;3组两次均免疫猪O型口蹄疫合成肽疫苗;4组两次均免疫PBS。首次免疫后0、14、42 d,检测猪血清中FMDV VP1结构蛋白抗体及细胞因子IL-2、IL-4、IL-10和IFNγ的水平;首次免疫后42 d,检测T淋巴细胞增殖水平。结果 2次免疫后,2组猪血清中FMDV VP1结构蛋白抗体水平、细胞因子IL-2、IL-4、IL-10和IFNγ水平及T淋巴细胞增殖率均显著升高。首次免疫后42 d,2组上述指标中除IL-2水平外,与3组和4组比较差异均有统计学意义(P<0.05或<0.01);除IL-2、IL-10和IFNγ水平外,与1组比较差异均有统计学意义(P<0.05或<0.01)。结论 Quil A对O型口蹄疫疫苗具有较好的免疫增强作用。 Objective To investigate the effect of Quil A as an immunoenhancer of foot-and-mouth disease(FMD) serotype O vaccine.Methods Piglets negative for antibody against FMD virus(FMDV) serotype O were randomly divided into four groups.The piglets in group 1 were primed with FMD serotype O DNA vaccine pVIRIL18P1 and boosted with recombinant fowlpox virus FMD vaccine,while those in group 2 were primed with Quil A + FMD serotype O DNA vaccine pVIRIL18P1 and boosted with recombinant fowlpox virus FMD vaccine.However,the piglets in group 3 were immunized twice with FMD serotype O synthetic peptide vaccine,while those in group 4 twice with PBS.The levels of FMDV VP1 structural protein antibody as well as IL-2,IL-4,IL-10 and IFNγ in sera of piglets were determined 0,14 and 42 d,and T lymphocyte proliferation level 42 d after the first immunization.Results After the second immunization,the FMDV VP1 structural protein antibody,IL-2,IL-4,IL-10 and IFNγ levels in sera as well as T lymphocyte proliferation level of piglets in group 2 increased significantly.Forty-two days after the first immunization,the above-mentioned indexes in group 2,except the IL-2 level,showed significant difference with those in groups 3 and 4(P〈0.05 or P〈0.01),and the indexes except IL-2,IL-10 and IFNγ levels showed significant difference with those in group 1(P〈0.05 or P〈0.01).Conclusion Quil A showed satisfactory immunopotentiation to FMD serotype O vaccine.
