摘要
目的探讨眼针疗法对腹泻型肠易激综合征(D-IBS)的治疗作用及机制。方法将40只Wistar大鼠随机分为4组,对照组、模型组、匹维溴铵组和眼针组,每组10只。采用慢性应激结合束缚方法建立D-IBS模型,眼针组采用眼针治疗,针刺下焦区、大肠区、肝区、脾区;匹维溴铵组用匹维溴铵灌胃治疗,两组均治疗7天。HE染色观察结肠组织病理学变化;实时定量PCR测定结肠组织神经激肽1(NK1)的mRNA表达变化;免疫组化法测定结肠组织NK1的蛋白表达变化。结果各组大鼠结肠组织病理学无差异。与对照组比较,模型组结肠组织中NK1的mRNA表达水平显著升高(P<0.01);与模型组比较,眼针组结肠组织中NK1的mRNA表达水平显著降低(P<0.01);与模型组比较,匹维溴铵组结肠组织中NK1的mR-NA表达水平无统计学意义(P>0.05)。NK1免疫反应阳性细胞在结肠黏膜层、腺腔内、肌间神经丛、黏膜下神经丛等部位。与对照组比较,模型组结肠组织中NK1的蛋白表达水平显著升高(P<0.05);与模型组比较,眼针组和匹维溴铵组结肠组织中NK1的蛋白表达水平显著降低(P<0.05)。结论眼针能够降低D-IBS模型大鼠结肠组织中NK1的表达,从而对D-IBS起到治疗作用。
Objective To explore the effect and functional mechanism of eye-acupuncture therapy on the diarrhea irritable bowel syndrome(D-IBS).Methods Forty Wistar rats were randomized into four groups,control group,model group,pinaverium bromide group,and eye-acupuncture group,with 10 in each.We used chronic stress in combination with binding to make D-IBS models.The eye-acupuncture group was treated with eye-acupuncture at the auricular points Lower-jiao,Large intestine,Liver,and Spleen,and the pinaverium bromide group was treated with pinaverium bromide for intragastric feeding,for 7 days.The histopathological changes of colon were observed by HE staining,the Neurokinin1(NK1) mRNA expression changes were detected by realtime-PCR,and the colic NK1 protein expression changes were determined by immunohistochemical method.Results The result of HE staining showed that there was no morphological changes of colic tissues.Compared with the control group,the NK1 mRNA expression of colic tissues in the model group was significantly increased(P〈0.01).Compared with the model group,the NK1 mRNA expression of colic tissues in the eye-acupuncture group was significantly decreased(P〈0.01).Compared with the model group,the NK1 mRNA expression of colic tissues in the pinaverium bromide group was without statistical significance(P〉0.05).NK1 immunoreaction positive cells were existing in the colonic mucous layer,lumen of gland,intermuscular plexus,and submucous plexus.Compared with the control group,the colic NK1 protein expression in the model group was significantly increased(P〈0.05);compared with the model group,the colic NK1 protein expression in the eye-acupuncture group and pinaverium bromide group was significantly decreased(P〈0.05).Conclusion Eye-acupuncture could effectively decrease the colic NK1 expression of D-IBS model rats,playing a therapeutic role in treatment of D-IBS.
