摘要
目的观察吸入光气诱导的急性肺损伤大鼠血清中血管生成素(ang)-2、ang-1和ang-1/ang-2比值变化及其与肺损伤指标的关系。方法84只大鼠随机分为13个光气染毒组(光气组)和1个空气对照组,每组6只。观察不同时间后,用酶联免疫吸附(ELISA)法检测血清中ang-1、ang-2水平及肺泡灌洗液(BALF)中白细胞计数、总蛋白浓度。结果随着时间延长,光气组大鼠ang-2、肺湿/干重比值、BALF中白细胞计数、总蛋白浓度呈升高趋势,ang-1有降低趋势,血清中ang-2/ang-1比值呈升高趋势。6、8、10、12、14、16、18、20、22、24、48h光气组ang-2/ang-1比值明显高于空气对照组,差异有统计学意义(P〈0.01,P〈0.05)。ang-2与肺脏湿/干重比值、BALF中总蛋白浓度、BALF中白细胞计数呈正相关(栅直分别为0.441、0.283和0.439),差异有统计学意义(P〈0.01)。ang-1与肺脏湿/干重比值、BALF中自细胞计数的相关系数分别为-0.286和-0.362,差异有统计学意义(P〈0.01)。ang-2/ang-1比值与肺脏湿/干重比值、BALF中总蛋白浓度、BALF白细胞计数呈正相关(r值分别为0.683、0.534、0.710),差异有统计学意义(P〈0.01)。结论光气染毒后,大鼠血清中ang-2/ang-1比值与肺脏损伤指标呈正相关,可能可以作为判定急性肺脏损伤严重程度的早期重要指标。
Objective To observe the relationship between the indicators of lung injury and the ratio of angiopoietin-2 (ang-2) to angiopoietin-1 (ang-1) in serum of rats with acute lung injury induced by phosgene. Methods Eighty four aduh male SD rats were randomly divided into 13 phosgene groups and 1 control group. Control group was exposed to fresh air for 5 minutes; the phosgene groups were exposed to phosgene ( 100% purity of 8.33 mg/L phosgene ) for 5 minutes. At different time points after exposure, the wet/dry weight ratio of mouse lungs was examined, the levels of ang-1 and ang-2 in serum of rats were detected with ELISA assay, and the white blood cell count and total protein levels in bronchoalveolar lavage fluid (BALF) of rats were measured. Results In 48 h after exposure,the serum ang-2, lung wet/dry weight ratio, BALF white blood cell count, protein concentration tended to increase with time, and the serum ang-1 tended to decrease with time. The ratios of ang-2 to ang-1 at 6, 8, 10, 12, 14, 16, 18, 20, 22, 24 and 48 h after exposure in phosgene groups were significantly higher than that in control group (P〈0.05 or P〈0.01). There were the positive correlations between ang-2 and lung wet/dry ratio, BALF white blood cell count or protein concentration (P〈O.01). There were the negative correlations between ang-1 and lung wet/dry ratio or BALF white blood cell count (P〈0.01). There were the positive correlations between the ratio of ang-2 to ang-1 and lung wet/dry ratio, BALF white blood cell count or protein concentration (P〈0.01). Conclusion After exposure of rats to phosgene, there is a positive correlation between the ratio of ang-2 to ang-1 and lung injury indicators, which may be an early indicator of the severity of phosgene-induced acute lung injury.
