鸭肠炎病毒gC糖蛋白胞外区优势抗原表位的筛选与鉴定 被引量:3

摘要 为筛选出鸭肠炎病毒(DEV)gC糖蛋白胞外区的优势抗原表位,本试验通过抗原表位作图法对DEV-gC基因进行分段克隆并构建重组表达载体,表达蛋白经纯化后进行Western blot分析。结果显示,经过4轮筛选,DEV-gC糖蛋白优势抗原表位为第73-88位氨基酸。该优势表位的发现为DEV-gC糖蛋白具体功能区的研究、诊断试剂及表位疫苗的研制奠定了物质基础。 Based on the method of epitope mapping,the extracellular region of gC glycoprotein of duck enteritis virus(DEV) was dissected into a set of overlapping fragments which were used to construct the recombinant expression vector,respectively.The expression proteins were purified to analyze the antigenicity by Western blot.The results showed that the dominant epitope region is located in the 73-88 aa region after four times screening.The results from this study lay a foundation for the research of DEV-gC glycoprotein's function and develop a new strategy for diagnostic reagents and epitope vaccin.
