艾炷灸对D-半乳糖衰老小鼠大脑组织抗氧化和学习记忆的影响 被引量:13

Effects of moxa cone moxibustion on learning and memory,oxidation resistance of cerebral tissues in senile mice induced by D-galactose

摘要 目的:观察艾炷灸对D-半乳糖衰老小鼠大脑组织抗氧化与学习记忆的影响。方法:3月龄昆明雄性小鼠72只,随机区组分为6组:生理组、模型组、艾炷灸"足三里、悬钟"(简称艾灸1组)、艾炷灸"关元、百会"(简称艾灸2组)、电针组、尼莫地平组,每组12只。模型组及干预治疗组,连续42d颈背部皮下注射D-半乳糖;生理组连续42d颈背部皮下注射等量生理盐水,造模第13d开始干预治疗,1次/隔日,共15次。水迷宫行为测试、脑组织匀浆一氧化氮(NO)含量、一氧化氮合酶(NOS)、总超氧化物歧化酶(TSOD)及铜锌-超氧化物歧化酶(CuZnSOD)的活力检测。结果:①模型组"潜伏期"较生理组、艾灸1、2组、尼莫地平组延长(P<0.01或P<0.05),模型组"原象比"、"跨台次数"较其他各组缩短、降低(P<0.01或P<0.05);②模型组脑组织NO含量、总一氧化氮合酶(TNOS)、诱导型一氧化氮合酶(iNOS)活力较其他5组显著升高(P<0.01),其他5组脑组织NO含量、TNOS、iNOS活力比较无明显差异。模型组脑组织TSOD、CuZnSOD活力较其他5组显著降低,其他5组脑组织TSOD活力无明显差异;电针组、尼莫地平组脑组织CuZnSOD活力低于艾灸2组、艾灸1组,差异有统计学意义(P<0.01,P<0.05),其余各组间比较无差异。结论:艾炷灸可以通过提高脑组织总SOD、CuZnSOD的活力,保护脑细胞免受NO自由基损害,提高学习记忆,延缓脑组织老化。 Objective:To observe the influence of moxa cone moxibustion on learning and memory ability,the oxidation resistance of changes on cerebral tissue of the senile mice induced by D-galactose.Methods:72 male mice(3 months old) were randomly and equally divided into the model,the normal sodium control,the moxa cone moxibustion 1(cupoint ST36 and GB39),the moxa cone moxibustion 2(acupoint DU20 and RN4),the electroacupunctue(acupoint ST36 and GB39),the Nimodipine medicated groups.The treatment and model groups were given with D-galactose injection for 42days,while injected the same dosage of normal sodium as other groups in the normal sodium control.From the 13th day on the first injection,the four treatment groups were treated for every other day for one month.Morris Water Maze Test(MWM) test each group again,the nitric oxide(NO) content,the nitric oxide synthase(NOS) and activities,the total superoxide dismutase(TSOD),and the CuZn-superoxide dismutas(CuZnSOD) activity of tissue homogenates of the cerebral cortex were assayed by using nitrate reductase method,chemico-chromatometry and hydroxylamine method respectively.Results:The significant difference was observed between the model group and the normal sodium control group and the four treatment groups on the escape latency of place navigation,the escape lantency of model group was prolonged,the spanning(original) platform frequency and the targeted quadrant time % of model group during the probe test was decreased(P0.01,P0.05) compared with other five groups.The NO content and the total superoxide dismutase(TNOS),the induced nitric oxide synthase(iNOS) activities of cerebral tissues in model group was higher than that in other five groups(P0.01).There was no difference between other five groups.The total superoxide dismutase(TSOD),the CuZn-superoxide dismutas(CuZnSOD) activities of cerebral tissues in model group was lower than that in other five groups(P0.01).There was no difference between five others on TSOD activity.The CuZnSOD activities in the Nimodipine medicated and the electroacupunctue were lower than that of the moxa cone moxibustion 1 and 2(P0.01,P0.05),there were no difference between others on CuZnSOD activity.Conclusion:Through improving the TSOD,CuZnSOD activities and avoiding the NO damage,the moxa cone moxibustion on ST36 and GB39,DU20 and RN4 can enhance the learning and memory function and reduce oxidative damage on cerebral tissue of the senile mice induced by D-galactose.
