芪蚣抗纤方对免疫损伤性肝纤维化大鼠TGF-β1/Smads信号通路影响 被引量:5

Effect of Qigong Kangxian Prescription on immunity-damaged liver fibrosis rat in TGF-β1/Smads signal pathway

摘要 目的:观察芪蚣抗纤方(QF)对免疫损伤性肝纤维化大鼠TGF-β1/Smads信号通路影响。方法:采用猪血清攻击法复制免疫损伤性肝纤维化大鼠模型;用双抗体夹心ABC-ELISA法测定血清LN、HA、TGF-β1含量;免疫组化法观察TGF-β1、Smad2 mRNA、Smad3 mRNA、Smad7 mRNA表达。观察QF抗肝纤维化的分子机制。结果:①血清学指标结果:与正常对照组比较,模型组大鼠血清TGF-β1、LN、HA含量明显升高(P<0.05或P<0.01)。经QF干预后,大鼠血清TGF-β1、LN、HA含量明显下降(P<0.01)。②免疫组织化学结果:模型组大鼠肝组织TGF-β1胞浆强阳性表达,Smad2棕色阳性着色由胞浆转至胞核,Smad7无阳性表达。QF干预组大鼠肝组织TGF-β1、Smad2、Smad3蛋白表达下调(P<0.01)。结论:QF通过调节TGF-β1/Smads信号通路,能保护和修复受损的肝细胞,抑制胶原纤维的形成,发挥抗肝纤维化作用。 Objective:To study the effect of Qigong Kangxian Prescription on immunity-damaged liver fibrosis rat in TGF-β1/Smads signal pathway.Methods:Models of liver fibrosis induced by immunologic injury with intra-peritoneal injection of porcine serum in rats.The contents of LN,HA,TGF-β1 of serum were detected with method of ABC-ELISA;The protein expression of TGF-β1,Smad2,Smad3 and Smad7 of liver tissue were detected with method of immunohistochemistry.Results:①Compared with the normal control group,contents of TGF-β1,LN,and HA in serum of model group increased significantly(P0.05 or P0.01).With the treatment of Qigong Kangxian Prescription,contents of TGF-β1,LN,and HA in serum lowered obviously(P0.01).②The results of immunohistochemistry indicated there was intense expression of TGF-β1 in cytoplasm,the expression of Smad2 in cytoplasm transferred into cellular nucleus,and the expression of Smad7 was negative in the model group.The expression of TGF-β1,Smad2,and Smad3 decreased(P0.01) in two dosages of Qigong Kangxian Prescription treatment groups,and above all,the effect was obvious in the Qigong Kangxian Prescription large-dosage group.Conclusion:Qigong Kangxian Prescription could regulate the Tran membrane signal transduction of TGF-β1/Smads for treating hepatic fibrosis.
