柞蚕滞育蛹与非滞育蛹蛋白质表达差异初步分析

摘要 柞蚕是以蛹滞育越冬的昆虫之一,研究滞育蛹及非滞育蛹的蛋白质种类及含量的变化规律,有利于其滞育机理的解明。通过SDS-PAGE获得滞育与非滞育雄性柞蚕蛹的蛋白质图谱在分子质量16~105 kD范围内均出现清晰的25条带,其中大小为79、54、48、29 kD的蛋白质表达量较高,79 kD的蛋白质为高丰度表达的大分子质量蛋白质;采用2D-PAGE技术分析,在滞育蛹蛋白质样品中检测到的总蛋白质斑点数为400个,非滞育蛹蛋白质样品中检测到的总蛋白质斑点数为619个,非滞育蛹蛋白质斑点明显增多,二者蛋白点匹配率仅为37%,蛋白质的等电点变化剧烈,差异性较大,匹配率较低。研究结果提示柞蚕蛹滞育过程中蛋白质的数量和种类发生了明显变化。 Chinese oak silkworm(Antheraea pernyi) is one of the insect species that hibernate in form of diapause pupa.Understanding the variation regularity of protein varieties and contents in diapause and non-diapause pupae would facilitate clarification of diapause mechanism.SDS-PAGE results showed that there were 25 clear protein bands isolated from both diapause and non-diapause male pupae of A.pernyi,among which proteins of 79,54,48,and 29 kD had high expression levels and the 79 kD protein was a high molecular weight protein abundantly expressed.Two dimension electrophoresis(2D-PAGE) results showed that the detected number of total protein spots from protein sample of diapauses pupae was 400,and that of non-diapause pupae was 619.There were obviously more protein spots from non-diapause pupae.The matching rate of protein spots between them was only 37%.Isoelectric points of these proteins were greatly varied,with big difference and low matching rate.These experimental results suggest that protein varieties and contents in A.pernyi pupae have remarkable changes in the process of diapause.
