与EST-SSR连锁的抗性候选基因潜在功能分析

摘要 利用课题组已开发的80个具有良好多态性的EST-SSR标记对已构建的遗传连锁图进行加密。结果表明,共有16个EST-SSR标记在父母本间具有良好的多态性,多态性标记频率为20%;后代群体基因型数据经χ2测验符合1∶1分离比例;经连锁分析,16个差异EST-SSR标记被整合到已构建的具有19个连锁群的分子遗传连锁图谱中,标记分布在7个连锁群,包括LG E、LG J、LG D1b、LG F、LG B2、LG O、LG C2,并分别获得了2个与大豆孢囊线虫病抗性相关和4个与大豆疫霉根腐病抗性相关候选基因连锁的功能标记。研究为实现分子辅助育种的功能标记直接选择奠定基础。 A total of 80 EST-SSR markers developed in previous study of our research group were used for encrypting a genetic linkage map constructed ever before.The results showed that a total of 16 polymorphic EST-SSR(polyphic frequence was 20%) fit 1:1 by χ2 test.These 16 EST-SSR marker was intergated into seven linkage map(LG E,LG J,LG D1b,LG F,LG B2,LG O,LG C2) of the objective genetic linkage map.Moreover,two and four candidate genes with resistance to soybean cyst nematodes and phytophthora root rot were obtained which were associated with six functional EST-SSR markers.
