Exenatide对高糖培养的视网膜神经节细胞的保护作用

摘要 目的:评价Exenatide对视神经保护作用,并初步探索其对视网膜神经节细胞(retinal ganglion cells,RGCs)的保护机制。方法:体外培养RGCs,免疫荧光检测RGCs是否表达胰升糖素样肽-1受体(glucagon-like peptide-1receptor,GLP-1R),采用高糖作为细胞凋亡诱导剂建立RGCs凋亡模型,并用不同浓度Exenatide加入细胞培养基中进行干预,用CCK-8检测细胞存活率。结果:免疫荧光结果显示,RGCs存在GLP-1R的表达。CCK-8检测结果显示终浓度为0.5~1μg/L时Exenatide均可促进RGCs存活,其中0.5μg/L浓度较低且已有明显保护作用(P<0.05)。结论:Exenatide注射液对高糖引起的RGCs生存抑制有一定保护作用,这种保护作用很可能通过GLP-1R介导的细胞内保护机制产生。 AIM:In present study,we evaluate the neuroprotective effect of Exenatide,and further investigate the underlying mechanism of Exenatide-induced retinal ganglion cells(RGCs) protection.METHODS:In vitro,expression of glucagon-like peptide-1 receptor(GLP-1R) in RGCs cells was detected by immunofluorescence and apoptotic models was built by using high-glucose as cell apoptotic inducer.Different concentrations of Exenatide as the intervention was added to medium and cell survival rate was evaluated by CCK-8 test.RESULTS:Immunofluorescence results showed that RGCs expressed GLP-1R.CCK-8 test results indicated that 0.5-1μg/L concentration of Exenatide could promote the survival of RGCs,and 0.5μg/L concentration of Exenatide which was the low concentration had favourable effect to protect RGCs(P0.05).CONCLUSION:Exenatide protects RGCs from survival inhibition induced by high-glucose.The protection may depend on GLP-1R induced cell protective mechanism.
