组胺受体拮抗剂对高致病性猪蓝耳病血液学的影响 被引量:3

摘要 为探索组胺(HA)受体拮抗剂对高致病性蓝耳病(PRRS)病毒感染患猪血液学的影响,揭示内源性组胺在高致病性猪蓝耳病发生发展过程中的部分作用,通过人工使仔猪感染高致病性蓝耳病,并相应注射扑尔敏、西咪替丁后第5天采用RT-PCR方法检测仔猪血液中的PRRSV,第7天采用ETDA抗凝管颈静脉采血,全自动血液分析仪测定血液指标。结果表明,1)攻毒后第5天均扩出1条约1 064 bp的特异性目的条带,表明攻毒成功;2)西咪替丁组、阳性组仔猪血液中白细胞总数、淋巴细胞数量、单核细胞数量极显著低于阴性组(P<0.01),而扑尔敏组均显著高于阳性组(P<0.05);3)阳性组仔猪血液中血红蛋白浓度、红细胞压积显著低于阴性组(P<0.05),西咪替丁组红细胞数显著高于阳性组;4)扑尔敏组、西咪替丁组仔猪血液中血小板含量、血小板压积极显著高于阴性组(P<0.01),阳性组血小板含量极显著高于阴性组(P<0.01),阳性组血小板压积显著高于阴性组(P<0.05)。说明,扑尔敏能抑制高致病性蓝耳病患猪血液中白细胞总数、白细胞中淋巴细胞数量和白细胞中单核细胞数量的下降,稳定红细胞相关指标的数量,具有增强机体免疫力的作用,但促进了血小板的升高,西咪替丁能维持高致病性蓝耳病患猪血液中红细胞相关指标的稳定。 The piglets artificially infected with porcine reproductive and respiratory syndrome were injected with two histamine receptor antagonists (Cimetidine and Chlorpheniramine) and then PRRSV in tested piglets was detected by RT-PCR after 5 d and the blood indexes were determined by an automatic blood analyzer after 7 d to study the effect of histamine receptor antagonist on hematology of porcine reproductive and respiratory syndrome and to reveal the part effect of endogenous histamine on occurrence and development of porcine reproductive and respiratory syndrome. The results showed that. 1) a specific band with 1 064 bp was amplified after 5 d, which indicates that PRRSV is successfully attacked; 2) WBC, LYM and MON of positive piglets and piglets injected with Cimetidine was very significantly lower than negative piglets but WBC, LYM and MON of piglets injected with Chlorpheniramine was significantly higher than positive piglets; 3) HCB and HCT of positive piglets was significantly lower than negative piglets and HCB of piglets injected with Cimetidine was significantly higher than positive piglets; 4) PLT and PCT of piglets injected with Chlorpheniramine or Cimetidine was very significantly higher than negative piglets, PLT of positive piglets was very significantly higher than negative piglets, PCT of positive piglets was significantly higher than negative piglets and PLT of piglets injected with Chlorpheniramine was maximum among four tested groups. Chlorpheniramine can effectively inhibit WBC, LYM and MON decrease of piglets infected with porcine reproductive and respiratory syndrome, stabilize related indexes of red blood cell and improve body immunity, but promote PLT increase. Cimetidine can stabilize related indexes of red blood cell in piglets infected with porcine reproductive and respiratory syndrome.
