摘要
对国内豢养的瓶鼻海豚进行了血样采集,利用微卫星分型技术对相关个体进行了亲子鉴定。共采集到65头太平洋瓶鼻海豚和11头南瓶鼻海豚的血液样品,其中包括1头太平洋瓶鼻海豚和1头南瓶鼻海豚亲子鉴定所需的样本。通过8%聚丙烯酰胺凝胶电泳对物种特异性引物的扩增产物进行分型,得到了13对扩增稳定且多态性高的引物。Cervus软件的分析结果表明,有11对多态性引物适用于这两种海豚的亲子鉴定,并且成功鉴定出了两对父子关系。
The bool samples were collected in 65 common bottlenose(Tursiops truncatus) and 11 Indo-Pacific bottlenose dolphin(T.aduncus) and analysed by microsatellite genotyping method.The PCR products were genotyped using 8% polyacrylamide gel electrophoresis on a Sequi-Gen GT System.Thirteen polymorphic microsatellite primer pairs were found,in which eleven primer pairs were suitable for the paternity testing according to the Cervus analysis.By means of these evelen primer pairs,sires of them were identified successfully.
