摘要
目的建立高效液相色谱法测定血浆中氨甲喋呤浓度的方法。方法以乙腈∶0.02mol/L磷酸水溶液(10∶90)为流动相,监测波长为302nm。结果在0~35.2μmol/L范围内呈良好线性关系,r=0.99995。日内及日间测得的RSD分别为0.62%,3.03%,平均回收率为99.6%。血清中最低检测浓度为10ng/ml。结论本法具有快速、准确,操作简便,流动相成本低等优点。
Objective To establish a method for the Determination of MTX in human serum by HPLC.Methods The mobile phase consisted of acetonitrile:0.02mol/Lphosphoric acid aqueos solution(10∶90).UV Detector was set at 302 nm.Results The good linearity was from 0~35.2 (μmol/L),r=0.99995.Within day and between days were 0.62% and 3.03% respectly.The average recovery was 99.6%.The detection of serum concentration was of 10ng/ml.Conclusion This method has advantages of fast,accurate,simple,low cost mobile phase.
