外源性D-核糖对大鼠心肌缺血再灌注损伤的保护作用 被引量:7

摘要 目的:探讨外源性D-核糖对大鼠心肌缺血再灌注损伤的保护作用及其机制。方法:24只SD大鼠随机分成假手术组、缺血再灌注I(schemia/reperfusionI,/R)组、D-核糖组。假手术组:只予以左冠状动脉前降支穿线不结扎I;/R组:左冠状动脉前降支结扎30 min,再灌注120 min;D-核糖组:左冠状动脉前降支穿线前30 min,给予尾静脉缓慢推注D-核糖,余操作同I/R组。各组术毕取结扎处远端左心室游离壁心肌组织备用。RT-PCR检测心肌中细胞因子诱导的中性粒细胞趋化物-1(Cytokine-in-duced neutrophil chemoattractant properties 1,CINC-1)mRNA,ELISA检测髓过氧化物酶(Myeloperoxidase,MPO),Western blot检测核因子-κB(Nuclear factor-κB,NF-κB),HE染色观察心肌病理改变。结果:与假手术组相比,D-核糖组I、/R组CINC-1mRNA的表达量,NF-κB和MPO的量均明显增高(P<0.05),心肌组织病理学变化明显加重,中性粒细胞浸润明显增多。与I/R组相比,D-核糖组CINC-1mRNA的表达量,NF-κB和MPO的量又明显减少(P<0.05),心肌组织病理学变化明显减轻,中性粒细胞浸润明显减少。结论:外源性D-核糖对SD大鼠心肌缺血再灌注损伤有保护作用,其作用机制可能与减少心肌组织中NF-κB的含量,降低心肌组织CINC-1mRNA的表达,减少心肌中MPO含量,减少心肌组织中性粒细胞浸润有关。 Objective:To investigate the effect and the mechanism of D-ribose on myocardial ischemia/reperfusion injury of rat hearts.Methods:24 SD rats were randomly divided into sham group,ischemia/reperfusion group(I/R group)and D-ribose group(n=8).The rats in sham group did not undergo the left anterior descending coronary artery(LAD)ligation;The rats in I/R group underwent the LAD ligation for 30 min,then were reperfused for 120 min;The rats in D-ribose group(D-ribose group)were administered D-ribose(800 mg/kg)through tail vein 30 min prior to the LAD ligation.The left ventricular samples of the three groups were prepared for investigation.The expression of CINC-1mRNA was detected by RT-PCR;myeloperoxidase(MPO)was detected by ELISA;nuclear factor-κB(NF-κB)was detected by Western blot,and cardiac HE sections were observed under light microscope.Results:Compared with sham group,higher expression of CINC-1 mRNA,NF-κB and MPO were detected in I/R group and D-ribose group(P〈0.05)with more remarkable histopathologic changes and neutrophil infiltration.Compared with I/R group,less expressions of CINC-1 mRNA,NF-κB and MPO were detected in D-ribose group(P〈0.05)with less remarkable histopathologic changes and neutrophil infiltration.Conclusion:Exogenous D-ribose might play protective effect on myocardial ischemia/reperfusion injury of rat hearts.The mechanism could be related to its decreasing the expression of NF-κB,CINC-1 mRNA and MPO as well as reducing neutrophil infiltration.
