摘要
目的观察对体外培养的骨髓基质细胞Hes1基因干扰后Mash1基因的表达变化。方法采用全骨髓培养法体外分离培养获得骨髓基质细胞,倒置显微镜下观察其形态变化,应用实时定量PCR技术分析在干扰Hes1基因表达的情况下Mash1基因表达的变化。结果应用200nM,100nM,50nM,25nM四种不同浓度梯度的Hes1的siRNA干扰抑制Hes1基因,与对照组相比Hes1表达均降低。200nM干扰后Hes1基因降低的程度是原来的0.036(P<0.05),50nM干扰后Hes1基因降低的程度是原来的0.076(P<0.05),优于其他浓度的干扰效果。Hes1降低可使Mash1基因表达增加,且Hes1降低越显著,Mash1表达增加越高。结论体外培养BMSCs过程中干扰Hes1基因对Mash1基因的表达有影响,且呈负相关调控关系。
Objective To explore the expression of Mash1 after the inhibition of the Hes1 gene in BMSCs.Methods BMSCs isolated from bone marrow were cultured in vitro.The morphologic feature and growth status of BMSCs and differentiated BMSCs were observed by inverted microscopy.The expression of Mash1 was analyzed by real-time quantitative PCR.Results The concentrations of Hes1 siRNA were 200nM,100nM,50nM or 25nM.The expression of Hes1 was decreased in comparison with that of the control group.The level of Hes1 at the concentration of 200nM in interfered BMSCs was 0.036-fold of that in control BMSCs(P0.05),and the level of Hes1 at 50nM in interfered BMSCs was 0.076-fold of that in control BMSCs(P0.05),both better than the others.The expression of the Mash1 gene was increased after the inhibition of the Hes1 gene in a dose-dependent manner.Conclusion Hes1 can down-regulate the expression of the Mash1 gene in BMSCs.
