摘要
目的:研究耐药性癫痫患者脑组织内依赖细胞周期的蛋白激酶13(Cell cycle-dependent protein kinase 13,CDK13)mRNA的表达。方法:在基因芯片扫描的基础上,运用逆转录-多聚酶链反应(Reverse transcription polymerase chain reaction,RT-PCR)技术检测CDK 13mRNA在难治性癫痢患者脑部的变化。结果:基因芯片扫描显示CDK13 mRNA在耐药性癫痫患者脑部比正常人表达上调2.159倍,RT-PCR检测同样显示CDK13 mRNA在耐药性癫痫患者脑部表达增强,信号明显增高,提示候选基因CDK-13 mRNA在耐药性癫痫患者脑组织中表达上调,RT-PCR与cDNA芯片结果一致。结论:虽然CDK13具体生物功能尚不明确,但我们认为CDK13可能作为重要的调节因子参与了神经元凋亡等分子事件。CDK13mRNA表达上调可能在耐药性癫痫发病机制中起一定作用。
Objective:To study the change of CDK13 mRNA expression in the brain tissue of patients with drug-resistant epilepsy so as to explore the possible mechanisms of the pathogenesis of refractory epilepsy.Methods: The microarray scanning and reverse transcription polymerase chain reaction(RT-PCR) were used to evaluate CDK-13mRNA expression in the brain tissue of 40 refractory epileptic patients and 8 normal controls.Results:As compared with the 8 normal controls,there appeared a significant increase in CDK13 mRNA expression in the patients with drug-resistant epilepsy,with the ratio of cy5/cy3 being 2.159 in microarray scanning(CDK13 mRNA expression in the patients with drug-resistant epilepsy was upregulated by 2.159 times compared with the normal controls.),and the results of RT-PCR were coincided well with those of the cDNA chip.Conclusion: Further study should be made to clarify the precise function of CDK13.CDK13 may take part in the molecular event in neuronal apoptosis as an important modulator.The up-regulation of CDK13 mRNA expression may play an important role in the epileptogenic mechanisms of drug-resistant epilepsy
