CGRP及IgE在特应性皮炎的表达及其与病情的相关性

Relationship between Expression of CGRP,IgE and Disease Severity in Patients with Atopic Dermatitis

目的:探讨特应性皮炎(atopic dermatitis,AD)皮损内降钙素基因相关肽(calcitonin gene relat-ed peptide,CGRP)的表达与患者血清中IgE表达及病情的相关性。方法:选定实验组AD患者22例,对照组20例,采用荧光定量聚合酶链反应法测定皮肤内CGRP mRNA的定量表达,酶联免疫吸附试验(ELISA)检测血清中免疫球蛋白E(immunoglobulin E,IgE)的水平,采用湿疹面积及严重度指数(EA-SI)评分评估患者病情,分析AD患者CGRP mRNA的定量表达与IgE的表达及EASI评分相关性。结果:实验组皮损内CGRP mRNA及血清中IgE表达明显高于对照组的表达(t=7.05,P<0.001;t=10.24,P<0.01);且两者均与EASI评分呈正相关(r=0.587,P=0.004;r=0.447,P=0.035)。而CGRP mRNA的定量表达与IgE表达无相关性(r=0.161,P=0.474)。结论:CGRP及IgE的过度表达可能参与了AD的发病,CGRP或IgE可作为判断AD患者病情严重程度的指标之一。

Objective：To investigate the relationship between expression of calcitonin gene re- lated peptide, immunoglobulin E and disease severity of patients with atopic dermatitis. Methods： Twenty-two adult patients with AD and 20 healthy controls were sequentially enrolled in this study. The serum level of IgE and mRNA expression level of CGRP was measured by ELISA and fluores- cent quantitative polymerase chain reaction, respectively. Severity of the disease was measured by eczema area and severity index. The correlation between expression of IgE, mRNA level of CGRP and EASI was analyzed. Results：Both expression level of CGRP mRNA and serum IgE level in AD were significantly higher than that in healthy control （ t = 7.05, P 〈 0. 001 ; t = 10. 24, P 〈 0. 01 ）. Significantly positive correlation in AD was found between expression of CGRP mRNA and EASI scores（r =0. 587, P =0. 004）, as well as between expression of IgE and EASI scores（r = 0. 447 ,P =0. 035 ）, but no correlation was found between expression level of CGRP mRNA and serum IgE level （r = 0. 161, P = 0. 474）. Conclusion： Our results suggest that expression of CGRP and IgE play an important role and could be a valuable factor in the assessment of disease severity.
