摘要
目的研究羽扇豆醇对人髓系K562、NB4、SHI-01及淋系Jurkat白血病细胞的增殖和凋亡作用。方法以不加药物处理为对照组,设置5个羽扇豆醇浓度梯度分别处理人髓系、淋系白血病细胞,采用MTT法检测细胞活力及增殖情况,流式细胞术检测细胞凋亡率。结果随着羽扇豆醇浓度的升高,细胞增殖明显受到抑制。当浓度达到150μmol/L时,4种细胞都呈现出明显的增殖抑制作用(均P<0.05);与对照组相比,羽扇豆醇组细胞凋亡率明显增加(P<0.05)。结论羽扇豆醇可显著抑制人髓系K562、NB4、SHI-01及淋系Jurkat白血病细胞增殖并诱导其凋亡。
Objective To study the effect of lupeol on proliferation and apoptosis of myeloid leukemia cell lines K562,NB4,SHI-01,lymphoid leukemia cell lines Jurkat.Method Cells were treated with lupeol in five different concentrations in vitro.Compared with cells dissolvent only,proliferation inhibition was detected by MTT colorimetric assay,and cell apoptosis was analyzed by flow cytometry.Result The proliferation of both myeloid and lymphoid leukemia cells was inhibited significantly,and even 100% inhibition was reached in concentration of 300 μmol/L.When the drug concentration reached 150 μmol/L,all the four type cells showed significant inhibition of proliferation,the difference in which was statistically significant(P0.05).Compared with the negative control group,Lupeol treated group showed a significant increase in cell apoptosis(P0.05).Conclusion Lupeol inhibits leukemia cell proliferation and induced cell apoptosis.This study provides evidence for the possible clinical application of lupeol in leukemia treatment.
