肺炎支原体ORF6区蛋白重组抗原制备及在血清学诊断中的初步应用

摘要 目的本文应用肺炎支原体ORF6区重组抗原建立酶联免疫吸附实验(ELISA),用于检测肺炎支原感染者IgM抗体。方法通过已构建的质粒表达重组蛋白,用电洗脱法纯化后建立ELISA试验,并检测临床标本224份,与国外2种商品化试剂盒检测结果对照,初步考核检测方法的敏感性和特异性。结果与进口试剂盒相比阳性率基本相符,统计分析无显著差异。结论研究建立的间接ELISA方法特性强、敏感度高,为试剂盒的研制与开发奠定了基础。 To establish ELISA experimental method for IgM antibody of Mycoplasma pneumonia infector,recombined ORF6 protein from Mycoplasma pneumonia were produced.The constructed plasmid was induced to express the corresponding protein,then the recombined protein was purified by electroelution and applied in indirect ELISA to detect 224 clinical samples.The results were compared with two imported commercialization kits to check the specificity and sensitivity of diagnosis method initially.The results suggested that the diagnosis method was specific and sensitive.There was an application prospect for the M.pneumoniae ELISA kit.
