七叶皂苷钠对油酸制备急性肺损伤大鼠模型的疗效观察 被引量:3

摘要 目的观察不同剂量及时间七叶皂苷钠对油酸制备急性肺损伤模型大鼠的疗效。方法急性肺损伤模型由鼠尾静脉缓慢注射油酸复制,模型成功后给七叶皂苷钠(简称七叶)组静脉注射不同剂量七叶,给甲强龙组静脉推注甲强龙,给空白对照组和模型组注射等量生理盐水。除七叶高、低剂量组在处理2h、6h后观察外,其他组均处理2h、6h和18h后观察。检测指标为PaO2、W/D、血浆和肺组织SOD及MDA。结果①2h及6h时PaO2及W/D:与空白对照组比,2h及6h,模型组PaO2明显降低、W/D明显升高。与模型组比,2h及6h,七叶各剂量组PaO2明显升高、W/D明显降低;6h时,甲强龙组PaO2明显升高。与甲强龙组比,2h时,七叶中、低剂量组PaO2明显升高;6h时,七叶各组PaO2明显升高。与七叶高剂量组比,2h时,七叶中剂量组PaO2明显升高;②2h及6h血浆、肺组织SOD和MDA:与空白对照组比,2h及6h,模型组血浆及肺组织SOD明显降低、MDA明显升高。与模型组比,2h,七叶高、中剂量组血浆SOD明显升高;6h,七叶各组及甲强龙组血浆SOD明显升高;2h,七叶高剂量组肺匀浆SOD明显升高;2h及6h,七叶高剂量组血浆、七叶各剂量组及甲强龙组肺组织MDA明显降低;2h,甲强龙组血浆MDA明显降低。③18h时PaO2、W/D、血浆及肺匀浆SOD、MDA:与空白对照组比,18h,模型组PaO2、血浆及肺匀浆SOD明显降低,W/D、血浆及肺匀浆MDA明显升高;与模型组比,七叶中剂量组和甲强龙组PaO2、血浆及肺匀浆SOD明显升高,W/D、血浆及肺匀浆MDA明显降低;与甲强龙组比,七叶组PaO2、W/D、血浆及肺匀浆SOD、MDA无明显变化。结论不同剂量七叶皂苷钠对急性肺损伤模型大鼠均有抗炎、抗渗出作用,其较小剂量更安全,作用持续而稳定。 Objective To observe the effects of sodium aescinate of different doses and time on acute lung injury(ALI) model of rats induced by oleate. Methods Rat model of ALI was made by intravenous injection of oleic acid (0. 1ml/kg). Thirty minutes after model making, rats in sodium aescinate groups were injected with 6mg/kg,2mg/kg and lmg/kg sodium aeseinate while that of 5mg/kg medron in medron group,with equal sodium chloride in normal control group and model group. Except that rats in aescinate high and low dose group were observed 2h,6h after administration, rats in other groups were all observed 2h,6h and 18h after treatment. Detection indexes were the arterial blood oxygen pressure ( PaO2 ) ,lung wet/dry weight ratio (W/D) ,level of plasma and lung tissue superoxide dismutase (SOD) and malondialdehyde (MDA). Results (1)PaO2 and W/D: Compared with normal control group,2h and 6h,PaOEof rats in model group decreased while W/D increased significantly. Compared with model group,2h and 6h,PaO2 in aescinate dose groups increased while W/D decreased significantly;6h,PaO2 in medron group increased significantly. Compared with medrat group,2h,PaO2 in aescinate medium and low dose group increased significantly; 6h,PaO2 in aescinate groups increased significantly. Compared with aescinate high dose group,2h, PaO2 in aescinate medium dose group increased significantly. (2)Plasma and lung tissue SOD and MDA: Compared with normal control group,2h and 6h,plasma and lung tissue SOD in model group decreased while MDA increased significantly. Compared with model group, 2b,plasma SOD in aeseinate high,medium group increased significantly;6h,plasma SOD in aeseinate groups and medron group increased significantly;2h,lung tissue SOD in aescinate high group increased significantly; 2h and 6h,plasma MDA in aescinate high group and lung tissue MDA in aescinate groups and medron group decreased significantly. (3)PaO2, W/D, plasma and lung tissue SOD, MDA:Compared with normal control group, 18h,PaO2. plasma and lung tissue SOD in model group decreased while W/D,plasma and lung tissue MDA increased significantly. Compared with model group, PaO2, plasma and lung tissue SOD in aescinate medium dose group and medron group increased while W/D,plasma and lung tissue MDA decreased significantly. Compared with medron group,there was no significant difference in PaO2 , W/D,plasma and lung tissue SOD, MDA in aeseinate medium dose group. Conclusion Different doses of sodium aescinate have anti -inflammatory and antiexudative effect on ALI model rats while the small dosage with more secure,sustainable and stable effectiveness.
