摘要
目的研究石菖蒲不同部位对戊四唑点燃癫痫模型大鼠皮质多药耐药基因(MDR)表达产物P-糖蛋白表达强度的影响。方法 SD大鼠80只,腹腔注射戊四唑(PTZ)溶液35mg/kg体质量,隔天1次,共14次。点燃成功的大鼠,分8组,每天灌胃1次,分别给予石菖蒲挥发油50mg/kg体质量、石菖蒲去油水提液高剂量28g/kg体质量、中剂量14g/kg体质量、低剂量7g/kg体质量、β-细辛醚100mg/kg体质量、α-细辛醚70mg/kg体质量,阳性对照组给予丙戊酸钠(VPA)126mg/kg体质量,模型组给予等体积生理盐水。另设正常组5只,正常喂养,不作任何处理。治疗36d后注射同剂量戊四唑点燃测试药效,取大脑皮质制成蜡块,免疫组织化学染色,200倍镜下观察神经元中多药耐药基因表达产物P-糖蛋白的表达强度。结果与正常组及阳性对照组(VPA)比较,石菖蒲去油水提液中、低剂量组大鼠皮质P-糖蛋白(P-gp)表达强度降低(P<0.05),β-细辛醚组大鼠皮质P-糖蛋白表达强度降低(P<0.01);与模型组比较,β-细辛醚组大鼠皮质P-gp表达强度降低(P<0.05);与α-细辛醚组、石菖蒲挥发油组、石菖蒲去挥发油水提液组高剂量组相比,β-细辛醚组P-gp表达强度降低(P<0.05)。结论β-细辛醚和石菖蒲去挥发油水提液中、低剂量组能降低戊四唑点燃大鼠皮质P-gp表达强度,以β-细辛醚作用最强。
Objective The research starts to explore the impact of different parts of Acorus Tatarinowii Schott pentylenetetrazol kindled rats on cortical P-glycoprotein expression of Multidrug resistance gene product.Methods 80 SD rats,intraperitoneal injection of pentylenetetrazol solution,35 mg/kg body weight,one time every other day,a total of 14 times.The successfully kindled rats are divided into 8 groups and drug is fed into the stomach once a day.Acorus tatarinowii volatile oil was given at 50 mg/kg body weight;high doses of Acorus tatarinowii liquid extract with the volatile oil removed at 28g/kg body weight;Medium doses were given at 14g/kg body weight;low-doses were given at 7g/kg body weight,β-asarone 100 mg/kg body weight,α-asarone 70mg/kg body weight.Positive control group was given sodium valproate(VPA) at 126mg/kg body weight treatment.The normal group of 5 rats was given a normal ration without any special treatment.The model group was given the same volume of physiological saline.36 days after treatment with the same dose of intraperitoneal injection of pentylenetetrazol,a test was done to determine the efficacy.All rats were decapitated and their brains were obtained,made of wax block to take the cerebral cortex.P-glycoprotein expression in cortex was measured by means of immunohistochemistry.Results After treatment,compared with the normal group and positive control group(VPA),cortical P-glycoprotein expression in rats of medium and low dose groups of Acorus Tatarinowii water extracts with oil removed is reduce(P0.05),β-asarone group is reduce(P0.01).Compared with the model group,cortical P-glycoprotein expression in rats of β-asarone group is reduce(P0.05).Compared with α-asarone group、 Acorus tatarinowii volatile oil group,high doses group of Acorus tatarinowii liquid extract with the volatile oil removed,cortical P-glycoprotein expression in rats of β-asarone group is reduce(P0.05).Conclusion Cortical P-glycoprotein expression in rats of β-asarone group and medium and low dose groups of Acorus Tatarinowii water extracts with oil removed is reduce.The best result is shown in the β-asarone group.
