复方银杏丹参颗粒对H9c2细胞氧化应激损伤的保护作用

摘要 目的观察复方银杏丹参颗粒(Compound Yinxing Danshen Granules,CYDG)对大鼠心肌细胞氧化应激损伤的保护作用。方法利用过氧化氢(hydrogen peroxide,H2O2)作为外源性自由基生成系统,体内模拟心肌细胞的脂质过氧化损伤,观察CYDG对H2O2致心肌细胞损伤的保护作用。结果 CYDG能明显提高细胞生存率,降低乳酸脱氢酶的漏出率,并可使细胞内丙二醛含量降低,超氧化物歧化酶和过氧化氢酶的活性升高,亦可减少细胞内活性氧的产生及提高血红素加氧酶蛋白表达。结论 CYDG对H2O2造成H9c2心肌细胞氧化应激损伤具有明显保护作用,提高心肌细胞的抗氧化能力。 Objective To observe the protective effects of Compound Yinxing Danshen Granules(CYDG),a traditional Chinese medicine prescription for activating blood circulation and removing blood stasis and obstacles from the collaterals,and relieving pain,against oxidative stress damage in rat H9c2 cells in vitro.Methods Hydrogen peroxide(H2O2) was used as exogenous free radical generating system to induce the lipid peroxidation damage of cardiac muscle cells.The protective effects of CYDG against H2O2-induced damage on cardiac muscle cells were observed.Results The method of methyl thiazolyl tetrazolium(MTT) staining showed that CYDG significantly increased the rate of cell viability.CYDG could increasethe activities of catalase and superoxide dismutase,and decrease the levels of lactate dehydrogenase and malondialdehyde in in vitro cultured H9c2 cells.dichlorofluorescein-diacetate(DCFH) labeling showed that CYDG increased the levels of reactive oxygen species(ROS) in H2O2-stressed H9c2 cells.Western blot showed that CYDG increased the expression of heme oxygenase-1(HO-1) in H2O2-stressed H9c2 cells.Conclusion CYDG could protect H9c2 myocardial cells from the damage induced by H2O2-oxidative stress,and increase the antioxidant capacity of cardiac muscle cells.
