摘要 [目的]探讨黑木耳多糖(Polysaccharides From Auricularia Auricula,PFAA)对体外高糖培养乳鼠心肌细胞凋亡的抑制作用及其机制。[方法]将Wistar乳鼠原代细胞随机分为4组,即空白对照组;高糖组;0.4 mg/mL PFAA组;0.8 mg/mL PFAA组。用荧光显微镜测定各组细胞凋亡情况,RT-PCR法检测bcl-2、caspase-3基因表达量的变化。[结果]经PFAA干预后,凋亡细胞明显减少,bcl-2表达增加,caspase-3表达减少。[结论]PFAA抑制体外高糖培养的乳鼠心肌的凋亡,其机制可能与抑制caspase-3、激活bcl-2表达有关。 [Objective]To investigate the effect of Polysaccharides from Auricularia auricula(PFAA)on glucose on the cardiomyocytes apopotosis in cultured neonate rat and its mechanism.[Methods]Primitive cardiomyocytes of wistar neonate rats were randomly divided into normal control group,the glucose group,0.4 mg/mL PFAA group,0.8 mg/mL PFAA group.Cell apoptosis was determined by fluorescence microscope,and the levels of bcl-2 and caspase-3 gene expression were determined by RT-PCR.[Results]PFAA could inhibited the apoptosis of cardiomyocytes of Wistar neonate rats and improve the level of bcl-2 gene expression and decrease the level of caspase-3 gene expression.[Conclusion]PFAA inhibited the apoptosis of cardio-myocytes of wistar neonate rats through down regulating bcl-2 gene expression and up regulating caspase-3 gene expression.
