摘要
目的研究川芎嗪在哮喘小鼠气道炎症和气道重塑中的作用。方法 Balb/c雄性小鼠39只,随机分为对照组、哮喘组、川芎嗪干预哮喘组、地塞米松干预哮喘组,每组10只(其中对照组9只)。建立哮喘小鼠模型,酶联免疫吸附法检测血清及肺灌洗液(BALF)中肿瘤坏死因子α(TNF-α)水平,同时计数BALF细胞总数、BALF沉渣和外周血涂片嗜酸细胞(EOS)百分比,光镜观察肺组织结构,Image-pro plus图像分析软件测量支气管壁厚度和平滑肌厚度。结果川芎嗪干预哮喘组小鼠BALF中细胞总数和EOS百分比、血液EOS百分比、小鼠血清及BALF的TNF-α水平均明显低于哮喘组小鼠,差异有统计学意义(P均<0.05)。川芎嗪干预哮喘组小鼠肺组织病理学改变及支气管壁厚度和平滑肌厚度均明显低于哮喘组小鼠。结论川芎嗪能改善哮喘气道炎症状态,抑制哮喘小鼠气道重塑。
Objective To investigate the mechanism of Ligustrazine on allergic airway inflammation and airway remodeling in asthmatic mice.Methods Thirty-nine healthy male BALB/c mice were randomly divided into 4 groups: the normal group(n = 9),the asthma model group(n = 10),the Ligustrazine group(n = 10),and the dexamethasone group(n = 10).The asthma model was established.The pathologic changes of lung tissue were observed.Counts of eosinophil(EOS)and total cell numbers in bronchoalveolar lavage fluid(BALF)were counted,the concentrations of TNF-α in BALF and serum were measured by ELISA.The thickness of airway smooth muscle and bronchial wall in lung tissue were measured.Results Compared with the asthma group,total cell number in BALF,EOS % in serum and BALF and the level of TNF-α in BALF and serum in the group treated with Ligustrazine were significantly decreased(P 〈0.05).Compared with asthma group,both the thickness of total bronchial wall and the smooth muscle in Ligustrazine treated group were significantly decreased(P 〈0.05).Conclusions Ligustrazine can reduce airway inflammation,and inhibit the remodeling of bronchi.
