摘要
目的观察舒郁宁心中药对慢性应激抑郁模型大鼠行为及海马形态结构的影响。方法 60只SD大鼠随机分为正常对照组、模型组、舒郁宁心中药高、中、低剂量组、氟西汀组。复制孤养加慢性轻度不可预见性的应激抑郁模型(21d),造模成功后进行21d药物干预。采用敞箱实验和糖水消耗实验观察大鼠行为学变化,采用大鼠脑组织切片HE染色观察大鼠海马形态结构的变化。结果模型组大鼠水平与垂直运动次数明显少于正常对照组,舒郁宁心汤高剂量组、氟西汀组大鼠敞箱实验得分明显优于模型组。观察HE染色切片可见慢性应激引起海马形态结构发生改变,舒郁宁心中药和氟西汀可对抗这种改变。结论舒郁宁心汤具有抗抑郁作用,能改善慢性应激对大鼠海马神经元细胞的损伤。
Objective:To investigate the effect of Shuyuningxin Decoction on behavior and morphology and structure of hippocampal of rats with depression.Methods:60 SD rats were randomly divided into the normal control group,the model group,the fluoxetine group,the high dose of Shuyuningxin Decoction group,the middle dose of Shuyuningxin Decoction group and the low dose of Shuyuningxin Decoction group.After being induced the chronic unpredicted mild stress depression model for 21 days,different therapies were used in the several groups.Behavior changes were observed the open-field experiment and the syrup consuming experiment.The morphology and the structure of hippocampal were detected by the HE staining.Results:Scores in horizontal and vertical motion of the model group were significantly lower than those of the control group.Compared with the high dose of Shuyuningxin Decoction group and the fluoxetine group,scores of the open-filed experiment were superior to those of the model group.Chronic unpredicted mild stress can change the morphology and structure of hippocampal of rats.Shuyuningxin Decoction and fluoxetine can ameliorate these changes.Conclusion:Shuyuningxin Decoction could improve the behavior scores of the rats with depression and ameliorate the damage of hippocampal induced by chronic unpredicted mild stress.
