银杏内酯B对大鼠胚胎中脑神经细胞生长的影响

摘要 目的:探讨银杏内酯B(ginkgolide B,GB)对正常大鼠胚胎中脑神经细胞及海人藻酸(kainic acid,KA)损伤的中脑神经细胞生长的影响。方法:实验大鼠分为对照组、阳性对照组(神经生长因子,NGF组)、银杏内酯B组、损伤组(海人藻酸组)、NGF+KA组、GB+KA组。采用MTT分析、形态学观察等方法,观察细胞存活率和集落分化率。结果:中脑神经细胞经72 h培养后,与对照组相比银杏内酯B不同浓度组细胞存活率明显增高(P<0.05,P<0.01)。海人藻酸组在3 d开始与对照组相比,细胞存活率明显下降(P<0.05);银杏内酯B可逆转海人藻酸降低细胞存活率的作用,且随着银杏内酯B剂量的增加,逆转作用增强(P<0.05)。银杏内酯B,NGF组细胞集落分化率与对照组相比明显升高(P<0.01);海人藻酸组细胞集落分化率明显下降(P<0.01);银杏内酯B可部分逆转海人藻酸的抑制作用。结论:银杏内酯B能促进中脑神经细胞的生长与分化,并能部分拮抗海人藻酸对中脑神经细胞的损伤。 Objective: To study the effects of ginkgolide B on the normal neurons of embryonic rat midbrain and on the embryonic rat midbrain neurons injured by kainic acid(KA).Methods: The experimental objects were divided into control,positive control(NGF),ginkgolide B(GB),damage group(KA),NGF+KA,GB +KA groups.The survival ratio and the colony differentiation ratio of embryonic rat midbrain neurons were estimated by MTT assay and microscopy respectively.Results: ①Survival ratio of midbrain nerve cells: compared with the control group,the survival ratio of embryonic rat midbrain neurons in ginkgolide B groups were increasing significantly when it was cultured for 72 hours(P0.05,P0.01).The cell survival ratio in kainic acid group was lower obviously than that of control group at days of three(P0.05).The nerve cells,exposed to kainic acid,showed characteristic change of injury,which could be relieved by the treatment of ginkgolide B,and ginkgolide B showed the dose-dependent manner(P0.05).②Colony differentiation ratio of embryonic rat midbrain neurons: compared with the control group,the colony differentiation ratio of ginkgolide B and NGF groups were increasing obviously(P0.01),and KA can inhibit the colony differentiation ratio of embryonic rat midbrain neurons sharply(P0.01).Ginkgolide B can partly prevent the embryonic rat midbrain cells from damage by kainic acid.Conclusion: Ginkgolide B can promote the development of embryonic rat midbrain neurons and prevent the nerve cells from damage by kainic acid.
