摘要
分别以聚乙二醇、明胶、海藻酸钙、壳聚糖/聚乙二醇和多聚L-赖氨五种材料作为包被物用不同方法对丝素蛋白进行预处理,制备5种丝素蛋白包被支架,并探讨它们对HeLa,293T和B16三种细胞生长的促进效果.结果表明,HeLa,293T和B16分别适合多聚L-赖氨酸、壳聚糖/聚乙二醇和海藻酸钙的丝素蛋白包被方法.在包被支架上不同细胞均呈扁平状、集落性附着在丝素蛋白纤维上.本实验获得的丝素蛋白5种包被支架增强了丝素蛋白的弹性、柔软性和黏附性,提高了丝素蛋白的可操作性,且对细胞具有较好的生物相容性和明显的促进生长作用.
Silk fibroin was coated by five materials(PEG,gelatin,calcium alginate,chitosan/PEG and poly-L-lysine) with different methods,and then five kinds of frames were prepared.Their growth-promoting effects of those frames on cells HeLa,293T and B16 were discussed.The results show that cells HeLa,293T and B16 were suitable for the coated silk fibroin by poly-L-lysine,chitosan/PEG and calcium alginate respectively.On the coated silk fibroin,different cells were flat,colony attached on silk fibroin.Those five coated frames we have got really enhanced the elasticity,softness and adhesion,and improved the operability of silk fibroin.Moreover those coated frames have good biocompatibility with an evidently promotional effect on cell proliferation.
