固相萃取-高效液相色谱法测定豆腐皮中碱性橙Ⅱ的含量 被引量:3

摘要 建立了一种高效液相色谱法测定豆腐皮中碱性橙II的方法。采用无水乙醇提取,强酸性阳离子固相萃取柱净化,以Kromasil 100-5C18柱(5μm,250 mm×4.6 mm)为分析柱,以甲醇+20 mmol/L乙酸铵溶液(体积比65∶35)为流动相,光电二极管阵列检测器波长449nm处检测。该方法的线性范围为0~5μg/mL,定量下限为0.01 mg/kg,回收率>80%,RSD为3%。经过液相色谱串联质谱进行方法确证,证明了方法的可靠性。 A method of detecting Basic Orange 2 in bean curd shin was established.Sample was extracted with ethanol.The extracts were depurated by Strata X-C column and analysed by Using Kromasil 100-5C18 Column,isocratic mobile phase including 10 mmol/L ammonium acetate and methanol.The amounts were quantified by liquid chromatography with PDA detector.The linear range was 0~5μg/mL and the relative standard deviation(RSD) was below 5%.Recoveries were greater than 80% and the limit of quantitation was 0.01mg/kg.The positive sample will be further confirmed by LC/MS/MS.
