摘要
目的:探讨调气消积汤对Lewis肺癌移植瘤血管生成的影响。方法:将48只C57BL/6小鼠常规接种Lewis肺癌,5天后选取荷瘤成功的小鼠随机分4组:模型组、调气消积汤组、顺铂组、综合组(调气消积汤+顺铂组),每组10只。每天分别予饮用水、调气消积汤各0.4ml灌胃共12天;隔日1次腹腔注射生理盐水/顺铂0.13ml,共3天。停药后24小时取材,应用WesternBlotting法检测Lewis肺癌小鼠肿瘤组织的微血管密度(MVD)、血管内皮生长因子(VEGF)和基质金属蛋白酶-9(MMP-9)蛋白的表达。结果:各治疗组的MVD、VEGF和MMP-9蛋白的表达均低于模型组,差异具有统计学意义(P<0.05或P<0.01);调气消积汤组MVD、VEGF和MMP-9蛋白的表达与顺铂组相比,差异无统计学意义(P>0.05)。结论:调气消积汤可抑制肿瘤血管形成达到抗肿瘤的作用。
Objective:To explore the effect of tiaoqi xiaoji decoction on angiogenesis of Lewis transplanted lung cancer mice.Methods:C57BL/6 mice were inoculated with Lewis lung cancer cells,the tumor-bearing mice were chosen to divided into four groups,model,tiaoqi xiaoji decoction,DDP and mixed(tiaoqi xiaoji decoction and DDP) groups,each group contained 10 mice.Each group was given the corresponding medicine(ig water or tiaoqi xiaoji decoction once a day for 12 days,ip NS or DDP once every two days for 3 days).The microvascular density(MVD),vascular endothelial growth factor(VEGF) and matrix metalloproteinases-9(MMP-9) were determined by Western blotting method.Results:The MVD,VEGF and MMP-9 expressions in the three treated groups were obviously lower than those in the model group,there were no differences between the tiaoqi xiaoji decoction group and DDP group on these indexes,the MVD and VEGF expressions in the mixed group were both the lowest.Conclusion:Tiaoqi xiaoji decoction could inhibit the angiogenesis of Lewis lung cancer by reducing the expressions of MVD,VEGF and MMP-9.
