GFAP及PKC在电磁脉冲辐照大鼠额叶皮层中的表达及意义

The expression and significance of GFAP and PKC in the rat frontal cortex after electromagnetic pulse exposure

摘要 目的探讨胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)及蛋白激酶C(protein kinase C,PKC)在电磁脉冲(electromagnetic pulse,EMP)辐照成年雄性SD大鼠额叶皮层中的含量变化及其意义。方法将成年雄性SD大鼠随机分为假辐照组(sham)和辐照组,辐照组又按辐照后取材时间不同分为0.5 h,1 h,3 h,6 h和12 h。采用免疫组化法分析EMP辐照后额叶皮层中GFAP表达情况,采用酶联免疫吸附实验(ELISA)分析额叶皮层中总PKC含量;选用Western Blot法分析PKC(PKC-α,PKC-βⅠ,PKC-βⅡ),PKC-α和PKC-βⅡ蛋白含量。结果 EMP辐照后额叶皮层中GFAP阳性表达细胞数增加,胞体增大,且峰值均出现在辐照后3 h(P<0.05)。额叶皮层中总PKC含量在EMP辐照后0.5 h增加且达到峰值(P<0.05),之后逐渐恢复,至12 h达到假辐照组水平;Western Blot结果显示PKC及PKC-βⅡ在EMP辐照后0.5 h和1 h表达量增加(P<0.05),之后恢复至假辐照组水平,PKC-α在辐照各组与假辐照相比差异均不具有统计学意义(P>0.05)。结论在EMP辐照引起大鼠额叶皮层损伤过程中,PKC含量增加可能起重要作用。 Objective To study the expression and significance of glial fibrillary acidic protein(GFAP) and protein kinase C(PKC) in the frontal cortex of adult male SD rats which were exposed to electromagnetic pulse(EMP).Methods Male SD rats were sham or whole-body exposed to EMP at 200 kV/m for 200 pulses(1 Hz).At 0.5 h,1 h,3 h,6 h,12 h after EMP exposure,GFAP was detected by immunohistochemistry method,total PKC content in the frontal cortex was estimated by enzyme-linked immunosorbent assay(ELISA),and the expressions of PKC(PKC-α,PKC-βⅠ,PKC-βⅡ),PKC-α and PKC-βⅡ were determined by Western Blot method.Results The positive cells of GFAP expression increased in the frontal cortex after EMP exposure and the peak was at 3 h after exposed EMP(P0.05).Total PKC content was increased at 0.5 h,1 h,3 h and 6 h after EMP irradiation compared with sham group(P0.05),and the peak was at 0.5 h group.It recovered to the level of sham group at 12 h.Western Blot showed that the expressions of PKC and PKC-βⅡ were increased at 0.5 h and 1 h after EMP exposure compared with sham group(P0.05).PKC-α was not significantly changed(P0.05).Conclusions EMP could cause the damage of the rat frontal cortex and PKC might play an important role in the procedure.
