摘要
研究藤黄酸季铵化壳聚糖纳米粒的主要药剂学性质。以天然聚合物壳聚糖为原料采用两步季铵化合成了季铵化壳聚糖(TMC),通过混合溶剂法制备了藤黄酸(gambogic acid,GA)TMC纳米粒(nanoparticles,NPs),用FT-IR和1HNMR表征TMC结构并计算出季铵化程度(DQ)并用FT-IR、1HNMR及透射电镜(TEM)表征载药纳米粒结构。NPs的载药量为(23.28±0.98)%,包封产率为(74.94±4.00)%,粒径为(98.24±3.5)nm。TEM观察载药纳米粒的形态为100 nm左右的球体。FT-IR和1HNMR表征证实GA被包埋在纳米粒的内部。用混合溶剂法可以成功制备得到藤黄酸季铵化壳聚糖纳米粒。
N-trimethyl chitosan(TMC) has been synthesized via two-step method using natural chitosan and characterized by FT-IR and 1HNMR,the degree of quaternization(DQ) was also obtained from the analysis of 1HNMR.TMC Nanoparticles(NPs) containing gambogic acid(GA) was prepared using mixed solvent method,the loading efficiency,entrapping efficiency and size of the NPs were(23.28±0.98)%,(74.94±4.00)% and(98.24±3.5) nm respectively.The structure of NPs containing GA was verified by FT-IR and 1HNMR,it’s demonstrated that GA was encapsuled into the inner core of the NPs.
