Semaphorin 5A(SEMA5A)基因中rs7702187位点的多态性与帕金森病遗传易感性关系的meta分析

摘要 目的运用meta分析的方法综合评价探讨Semaphorin 5A(SEMA5A)基因中rs7702187位点的多态性与帕金森病(PD)易感性的关系。方法检索PubMed、Medline和Embase,纳入内容涉及SEMA5A基因中rs7702187的多态性与帕金森病易感性的独立病例对照研究。各文献符合研究方法相似和有综合的统计指标等纳入标准,语种不限,排除不符合纳入标准和未涉及SEMA5A基因中rs7702187位点的多态性的对照研究。应用RevMan4.2软件进行统计分析。结果 7篇文章共12组不同种族人群纳入分析,共有PD组3 539例,对照组3 250例。在西方人群中,SEMA5A基因rs7702187位点的AA基因型分析:OR=0.99,95%CI=[0.69,1.43],P=0.96;A等位基因分析:OR=0.87,95%CI=[0.79,0.96],P=0.008;T等位基因分析:OR=1.02,95%CI=[0.82,1.27],P=0.88。在东方人群中,AA基因型分析:OR=1.05,95%CI=[0.93,1.52],P=0.16;A等位基因分析:OR=1.05,95%CI=[0.93,1.52],P=0.16;T等位基因分析:OR=0.84,95%CI=[0.70,1.00],P=0.05。结论在东方人群中,SEMA5A基因rs7702187位点的多态性与PD的易感性无关联。在西方人群中,SEMA5A基因rs7702187位点的A等位基因与PD的易感性有关,携带A等位基因的人群患PD的风险增加。 【Objective】 To investigate the relationship of Semaphorin 5A(SEMA5A) and genetic risk of Parkinson's disease.【Methods】 By searching Medline,PubMed,Embase,and Web of Science(updated to September 31,2010),articles associated with the relationship of SEMA5A and risk of Parkinson's disease were collected.All studies selected should meet all of the inclusion criteria such as "case-control studies","similar study method ","sufficient published data for estimating an odds ratio(OR) with 95% confidence interval(CI)".Studies meeting exclusion criteria were removed.【Results】 12 studies about different populations including 3539 cases and 3250 controls were selected.In eastern subgroup,there was no difference in the frequence of AA genotype,A allele and T allele between the PD patients and the controls(AA genotype: OR=1.05,95%CI=[0.93,1.52],P =0.16;A allele: OR=1.05,95%CI=[0.93,1.52],P =0.16;T allele: OR=0.84,95%CI=[0.70,1.00],P =0.05).In western subgroup,people carrying a A allele had an OR of 0.87(P =0.20 for heterogeneity,95%CI=[0.79,0.96]).But the frequence of AA genotype and T allele of western origin showed no significantly difference between PD group and control group(OR=0.99,95%CI=[0.69,1.43],P =0.96;OR=1.02,95%CI=[0.82,1.27],PP =0.88).【Conclusion】 This meta-analysis suggests that there is no association between SEMA5A rs7702187 polymorphism and PD in east population,but SEMA5A gene rs1102187 polymorphism may be associated with PD disease in west population.The SEMA5A rs7702187 A allele may be a genetic risk factor for developing PD in west population.
