低氧诱导因子1α在白蛋白过负荷致肾小管上皮细胞转分化中的作用

摘要
长期蛋白尿可致肾脏纤维化,细胞外基质(ECM)的主要来源之一是转分化的肾小管上皮细胞(TEC),因此上皮细胞-间充质细胞转分化(EMT)是蛋白尿患者肾脏纤维化的主要机制之一。低氧诱导因子1(HIF-1)可诱导多种细胞转分化。本研究检测大鼠TEC(NRK-52E细胞)在含高浓度牛血清内蛋白(BSA)的培养基中是否发生转分化,以及shRNA-HIF—1α能否抑制BSA致NRK-52E细胞转分化的作用,以探讨HIF—1α在蛋白尿致肾脏纤维化中的作用。
