Expression of Apelin/APJ in Polycystic Ovary of Model Rats

摘要 目的:探讨Apelin/APJ在多囊卵巢综合征(PCOS)模型大鼠卵巢组织中的表达。方法:21日龄SD大鼠肌肉注射脱氢表雄酮(DHEA),建立PCOS大鼠模型(实验组,n=15),并以等量溶剂油代替DHEA为对照组(n=15),观察卵巢相对重量;HE染色观察卵巢改变;测定E2、T、P、FSH、LH水平;采用免疫组织化学法观察Apelin/APJ在PCOS卵巢中的分布与表达;荧光定量RT-PCR(quantitative RT-PCR)检测Apelin/APJ mRNA表达,Western-blotting检测Apelin/APJ蛋白表达。结果:apelin/APJ分别表达于大鼠卵泡膜细胞、颗粒细胞;实验组卵泡内膜、颗粒细胞中Apelin/APJ mRNA及蛋白表达均高于对照组(P<0.05)。结论:apelin/APJ的异常表达可能与PCOS复杂的发病有关。 Objective: To explore the role of apelin and APJ in the pathogenesis of PCOS.Methods: Female SD rats aged 21 d were divided into two groups in pairs.The rats of exprimental group(n=15) were intramuscular injected each day with dehydroeplandrosterone(DHEA)(60 mg/kg,qd×21 d,i.m.) and the rats of control group(n=15) injected with oil(0.2 ml);Ovarian weight and sex hormone levels in rat blood of both groups were determined;Ovarian change was observed by HE staining.The expression of apelin and APJ were detected by immunohistochemical staining in the rat's ovaries;The levels of apelin and APJ mRNA and protein in ovary tissue were measured by quantitative RT-PCR and Western blotting technique.Results: Immunostaining of apelin and APJ in the granulosa cells and theca cells of PCOS rat's ovaries were obviously stronger than those of the control(P0.05,respectively).The levels of apelin and APJ mRNA and protein in ovary of PCOS were obviously higher than those in the control(P0.05,respectively).Conclusion: The disruption of apelin and APJ system may be involved in the pathogenesis of PCOS.
