摘要
目的:探讨寡糖sLeX与L-选择素(L-selectin)作为黏附分子在母-胎之间的识别与黏附的作用。方法:通过免疫荧光方法检测胚胎细胞(JAR细胞)与子宫内膜细胞(RL95-2细胞)的sLeX与L-选择素的表达,通过转染α1,3-岩藻糖基转移酶VII(fucosyltransferase VII,FUT7)过表达载体至JAR细胞和RL95-2细胞,调节细胞表面sLeX的表达,以及通过sLeX/L-选择素抗体封闭细胞,计算不同处理后JAR细胞与RL95-2细胞黏附率。结果:①胚胎细胞与子宫内膜细胞均表达sLeX和L-选择素;②sLeX抗体及L-选择素抗体分别封闭JAR细胞与RL95-2细胞后,JAR细胞与RL95-2细胞的黏附率下降;③FUT7过表达增加sLeX寡糖抗原的合成,促进JAR细胞与RL95-2细胞的黏附率。结论:sLeX/L-选择素组成的黏附系统在胚胎与子宫内膜的识别与黏附过程中发挥重要作用。
Objective: To explore the role of sLeX/L-selectin in the recognition and adhesion of embryo and uterine.Methods: The expression of sLeX/L-selectin on JAR and RL95-2 cells were detected by indirect immunofluorescence analysis.Overexpression of fucosyltransferase VII(FUT7) trafected RL95-2 cells and JARcells to increare the expression of sLex.Antibody blocking experiment was used to block thee sLeX/L-selectin.Then the adhesion percent of JAR cells to RL95-2 monolayer was calculated.Results: 1) sLeX and L-selectin co-existing in JAR and RL95-2 cells.2) After JAR cells and RL95-2 cells were blocked by sLeX or L-selectin antibody respectively,the adhesion percent of JAR cells to RL95-2 monolayers decreased.3) Elevated sLeX by FUT7 gene transfection enhanced adhesion of JAR cells to RL95-2 cell monolayer.Conclusion: sLex/L-selectin adhesion system plays an important role in the recognition and adhesion of embryo and uterine.
