竹叶叶绿体色素的分离与鉴定 被引量:7

摘要 研究乙醇、溶剂用量、提取温度、提取时间等因素对提取竹叶叶绿体色素的影响,通过正交试验确定提取竹叶叶绿体色素的最佳条件,进一步用薄层层析法及紫外光谱分离、鉴定叶绿素a、叶绿素b、β-胡萝卜素和叶黄素4种叶绿体色素。结果表明,以乙醇为溶剂,竹叶叶绿体色素的最佳提取条件为溶剂80%、溶剂用量为15∶1(V/V)、提取温度60℃、提取时间3.5 h,4种叶绿体色素的含量分别为占竹叶的0.37%、0.12%、0.083%和0.011%。 This experiment through researching the factors of influence to extract the chloroplast pigment from bamboo leaves,such as the solvent concentration,the solvent volume,the extracting temperature and time,and preliminarily determined the better technology conditions of preparing chloroplast pigment from bamboo leaves,and through an orthogonal test for choosing the best conditions of preparing chloroplast pigment paste from bamboo leaves.The results indicated that,which uses ethanol to be the extracting solvent,the conditions were as follows,solvent concentration 80%,the ratio of material to solvent 1∶15(V/V),extracting temperature 60℃,extracting time 3.5 h.Further used the Thin-Layer chromatography separate,and the UV/V was spectra authenticate chlorophyll a,chlorophyll b,beta carotene and xanthophyll,and measurement four kinds of the chloroplast pigment content of the difference for 0.37%,0.12%,0.083% and 0.011% of having the bamboo leaves.
