摘要
研究PLIN基因外显子2和内含子2多态性与肉鸭部分产肉性状的关系。以361只北京鸭(Z2系、Z4系、Z2×Z4杂交系)和樱桃谷鸭为材料,利用PCR-RFLP方法和测序技术,对PLIN基因外显子2和内含子2进行SNPs位点检测,并对SNPs位点与部分产肉性状进行关联性分析。结果显示,在外显子2检测到C→T突变,分析表明,樱桃谷鸭群体中,胴体质量、全净膛质量和腹脂质量分数在CC和CT型之间差异极显著(P<0.01),腹脂质量和皮脂质量分数在CC和CT型之间差异显著(P<0.05);北京鸭Z4系群体中,全净膛质量在TT型与CC、CT型之间差异显著(P<0.05);北京鸭杂交系群体中,全净膛质量在CC和CT型之间差异显著(P<0.05)。在内含子2检测到C→T突变,分析表明,北京鸭Z4系群体中,全净膛质量在EE和DE型之间差异极显著(P<0.01),EE和DD型之间差异显著(P<0.05);北京鸭杂交系群体中,胸肌质量在DD和DE型之间差异极显著(P<0.01),皮脂质量分数在DD和EE型之间差异显著(P<0.05)。说明PLIN基因单核苷酸多态性对肉鸭胴体、脂肪性状有一定的影响,可作为影响胴体和肉质的候选基因,指导优良肉鸭品种的选育工作。
Polymorphisms of PLIN gene exon 2 and intron 2 and their correlation with part of duck meat production traits were studied in this paper.361 Beijing ducks(Z2 strain,Z4 strain,Z2 × Z4 hybrid strain) and Cherry Valley ducks were used as materials and PCR-RFLP and sequencing methods were used for SNPs detection of exon 2 and intron 2 of PLIN gene.Correlation analysis was carried on between the discovered SNPs and part of the meat production traits in ducks.The SNP detection showed a nucleotide mutation(C→T) exists in exon 2 and the result of correlation analysis showed as follows: in Cherry Valley ducks,carcass mass,eviscerated mass and abdominal fat percentage were extremely different between the genotype CC and genotype CT(P〈0.01),and abdominal fat mass and sebum rate were significantly different between CC and CT(P〈0.05).In the Beijing duck Z4,there was a significant difference in eviscerated mass among genotype TT,CC,and CT(P〈0.05).In Beijing duck(Z2 × Z4),there was a significant difference between CC and TT(P〈0.05) in eviscerated mass and a nucleotide mutation(C→T) was detected in intron 2.The analysis showed in the Beijing duck Z4,eviscerated mass were extremely different between EE and DE genotypes(P〈0.01)and significantly different between EE and DD(P〈0.05).In the Beijing duck(Z2 × Z4),breast muscle mass between DD and DE was extremely different(P〈0.01) and sebum rate was significantly different between DD and EE(P〈0.05).This study indicated single nucleotide polymorphisms of PLIN gene have a certain influence on the duck carcass and adipose traits,thus it can be a candidate gene affecting carcass and meat quality,guiding the breeding work of good duck species.
